    Very nice presentation. A few items to add: Since the blank represents 0 concentration and 0 absorbance, you can plot that as a sixth data point (0,0). In fact, that is why the best-fit straight line (as yours appears to) should begin at the point where the axes meet. Also, I would point out that making sure the best-fit line leaves about as many data points above it as below it (as you did) ensures that this line is "averaging out" any imperfections in the measurements.

    It would, but for this particular class we are simply introducing the concept of how protein concentration and absorption relate to one another, as well as how a standard curve could be constructed from that obtained data. Beyond that, many laboratories still do perform standard curves alongside protein assays, so this is intended to provide the students with the protocol associated with those assays.

    Thank you for this video! I been trying to solve this through Beer's Law formula and only lead me to deeper confusion. Plotting my points and finding the X-axis with absorbance number is much easier.

    Help with a question ????? A compound absorb light in 227 nm. The standard curve calibration for the compound is Abs = 372x + 0,6432 which the concentration is g/100mL. The technique was analytical standard adding from fluoxetine hydrochloride (C17H18F3NO.HCl - molar mass = 345,79 g/mol). So determine the concentration in mol/L from the analysed solution. Show all calcs progression.

    Thanks and well explained in conceptual term. I would like to ask that if possible, please show the use of spectrophotometer because new researchers faced tedious problems particularly when no Lab technicians are available.

    Wouldn't it be more precise to use the beer-Lambert law equation to figure out the molar extinction coefficient of the Protien, then use that to find the unknown concentration?

    If I have a standard solution of 5.0 ug/ml P and want to make dilutions of 0.5, 1, 1.5, 2, 2.5, 3, 3.5, 4, 4.5, and 5 mg/L to make a standard curve to determine mg/L P in a sample how would I prepare those dilutions ?
