As far as I understand, Mark was informed just before shooting the scene, so that he could react accordingly (David Prowse apparently said the line "Obi-Wan killed your father" during the take) the only people who knew the twist at that point were Mark Hamill, George Lucas and the director Irvin Kerschner. The only other person who knew before the premiere was James Earl Jones who had to do the voice over with the real line, and maybe some sound engineer I suppose. Amazing that they managed to keep it secret even from the rest of the cast and everyone right up until the premiere.
It's a cheap shot, he learned from Obi-Wan. Whi used it again in the cantina, on Tattooine, Then Luke used it twice, one in the snow cave on Hoth, and again in the saber battle with Vader. Just before Vader yeeted Palpatine . 😂
Best part is this line wasn't actually in the shooting script. Lucas had Jones record it in secret. The original line was that Obi-Wan killed Luke's father. That's why Mark's response is so emotional.

I've always believed this is the exact moment where Vader shows and Luke feels that there is still some good left in him. It's the first time you see Vader actually show some compassion by telling Luke that he doesn't want to kill him. I've heard people say that he didn't want to kill Luke because he was his son but this is the same person who slaughtered all the younglings and Jedi without showing an ounce of remorse.
As an Adult, what I love about this scene now is watching videos here of kids seeing it for the first time and having the same reaction I had in the theater. It's such a powerful revelation that elicits such a human response. See we're not all that different and here's another example! Please people; keep making those videos!

When the line came up, all I felt was a sense of crushing disappointment. Then the film just ended with Han shrink wrapped and some Johnny-Come-Lately going off to rescue him.

Some of us really needed it definitely confirmed because Empire blew us away. And then Yoda finally said it, “Your father he is.” And then the big finish: Luke brought back the good in him. Vader became Anakin once again (we didn’t know anything about “the chosen one” yet-this was a son-saves-father father-saves son story). It was an amazing trilogy, even with the Ewoks, who eventually grew on us, and everything was fine and good.
I was 11 when I saw TESB in cinema for the 1st, still remember being shocked by the reveal. Even now 44 years on I can watch TESB all the way without skipping any scenes. BTW. It was No Luke, I am Your Father!
After watching it, I got unstoppable will to watch Star Wars in original voices! Firstly, there is no hard vocabulary, secondly, what a beautiful playing with voices, how many emotions, how many senses! So, I know I'll do this weekend)

George Lucas sat down with Mark and told him to do whatever it took to stay quiet about this specific scene, even with fellow cast members. After the crew watched the final product, Harrison Ford (Han Solo) turned to Mark and said "You didn't say that was going to happen" or something like that.

I'll be honest, as genuinely iconic and cool as this scene still is nearly 44 years later, I kinda feel it may have overstayed it's welcome a bit considering how heavily it's been referenced in pop culture over the years and because we, most people anyway, pretty much know the vast majority of the franchise lore, so it doesn't feel as impactful because of how Vader being Anakin has already been fully explored over the years. That said, this scene still rocks and Vader is still one of the best characters in fiction.

The timing for this scene is really important. It being cut and sped up really takes the magic out of it.
