  • DNA Fingerprinting Steps :-
    1. DNA is Isolated and extracted from the cell or tissue by centrifugation.
    2. DNA is cut into small fragments by treating with restriction enzyme.
    3. DNA fragments are seprated by agarose gel electrophoresis.
    4. The seprated DNA fragments by alkaline solution visualised under U.V radiation .
    5. Southern Blotting :- The DNA transferred from electrophoresis plate to nitrocellulose or nylon membrane sheet.
    6. VNTR probes are now added which bind to specific nucleotide sequence that are complementry to them . This is called Hybridisation.
    7. The hybridised DNA fragments are detected by Autoradiography. They are observed as Dark bands on X - ray film .
    8. These bands being of different sizes, give a characteristics pattern for an individual DNA .

      Digesting the DNA with the help of restriction endonuclease enzymes. Separating the digested fragments as per the fragment size by the process of electrophoresis. Blotting the separated fragments onto synthetic membranes like nylon. Hybridising the fragments using labelled VNTR probes.
      Samples collected from unidentified bodies can include: blood, buccal swabs, hairs, bone, teeth, fingernails, tissues from internal organs (including brain), muscle, and skin.
      DNA sample means a tissue, fluid, or other bodily sample of an individual on which a DNA analysis can be carried out.

      probe is a single-stranded sequence of DNA or RNA used to search for its complementary sequence in a sample genome. The probe is placed into contact with the sample under conditions that allow the probe sequence to hybridize with its complementary sequence.
