    what happens if it's cleaved by an enzyme with an insert of 500 bp?

      Hello. If I understand you correctly, what would happen if you added a 500 bp insert? Well for whatever the size of the plasmid is (lets say it is 1kb) you would see an increase in plasmid size, which would make the band run further up on the gel (around 1.5kb). We often use gel electrophoresis to confirm an insert in this way.
