How secondary antibody can bind to primary antibody ? As antigen and antibody can interact due to epitope and paratope present on the surface . Are there any specific structure on pri and sec.. antibody
In direct elisa we detect antigen from blood serum, here in indirect elisa we detect antibody from blood serum...agar antibodies hume known nhi hai ki patient serum me hai ta nhi..to use kaise detect kroge??? Aap socho..kuch detection method hona chahiye na..ya to hum tagged antigen use kre ya tagged antibody...tagged antigen aate nhi hai...isliye antibody detect krne ke liye tagged antibody use ki jati hai

The antibody extracted from human body and introduced in rat act as antigen for rat and rat body start producing antibody against it and called secondary antibody.
If there is no disease specific primary antibodies n all are wash out. after adding secondary antibodies why should not they bind to this antigens. Why secondary antibodies bind specifically only primary antibodies. Whether primary antibodies are unknown then how secondary antibodies are specific for primary antibodies n available in market. primary antibodies are not unknown...there is already so much of research work has been done for known diseases..for example we know which type of antibodies releases in malaria disease... or which type of antibodies releases in HIV disease...so we suspect... on the basis of suspection we choose secondary antibody..if color develops then patient positive if not develope then patient is negative
Okay, thank you but what would happen if a patient has the symptoms of some disease and the body is not producing the antibodies and when there is no antibody the substrate won't bind to it and we will not see the color and we call that patient a negative. What test we use to see the disease when a person's body is not making the antibodies.
