- In this video we discuss Sandwich ELISA in Hindi, ELISA Basics and ELISA Test procedure as well.
Sandwich ELISA typically require the use of matched antibody pairs, where each antibody is specific for a different, non-overlapping part (epitope) of the antigen molecule. A first antibody (known as capture antibody) is coated to the wells. The sample solution is then added to the well. A second antibody (known as detection antibody) follows this step in order to measure the concentration of the sample.

Nice explanation. You have mentioned epitope simply as they are present at the surface of pathogen. That's for sure it's a part of antigen. But I think more precisely it should be defined as the part of a antigen that binds to paratope of an antibody. Paratope is the part of Fab region of antibody. More precisely the Complementarity Determining Regions i.e. CDRs.
But yeah it's a very nice explanation.

Nice explanation ma'am. I have a question like for negative case you said that before adding substrate we do washing step but if the capture antibody and secondary antibody has bound to antigen but we don't know that as solution is transparent so at that time we also do washing or not like you didn't mentioned washing step in positive case before adding substrate like logically we should do washing. Can you please explain this?
Mam I have a question, so what we can do is we can link enzyme with the capture Abs only why to complicate the whole process? Then also the color reaction can happen as enzyme will be linkedin to the capture antibody only.

Generally an antigen has several or many different epitopes

In case of indirect detection of sandwich ELISA, the primary antibody used is derived from the patient or designed ?
The antibodies used in either direct & indirect sandwich method are genetically modified for particular antigen, Also this method is for antigen detection so only antigens are taken from patient's serum not antibodies.
