leprae is a gram positive so how can you use acid-fast stain to detect it?
Based on its cell wall structure, Mycobacterium leprae is a gram-positive, rod-shaped bacteria with a thick waxy coating. Due to this coating with mycolic acid, it cannot be stained very well with the regular Gram stain procedure. That is why we use the acid fast stain on it instead.

Why is acid alcohol used as a decolorizer in an acid fast stain, as opposed to a neutral alcohol solution?
because carbolfuchsin is a basic dye that is soluble in acidic solutions (like acid alcohol or the mycolic acids in the acid-fast cell walls)

I think you should go back and redo some of these videos as I feel it's important to mention what the mordant is for example.
so during an acid fast stain what color would the gram negative stain if a non acid alcohol were to be used as a decolorizer? clear?
probably red - without acid in the decolorizer, the carbol fuchsin would not leave the cells.

Why is ZN stain used instead of Gram stain in spite of the organism being a bacteria?
Acid-fast bacteria don’t stain well with Gram stain because of the waxy mycolic acids in their cell wall. So another stain is necessary.

hi . i have question. why do some lab use malachite green as a secondary stain ???
Personal preference, I guess.
Our lab did whenever we ran out of Methylene blue, lol. I think under certain microscopes Malachite green showed up better.
What outcome (color) is expected for a Gram positive, non-acid fast bacterium like Staphylococcus aureus if alcohol is mistakenly used instead of acid alcohol?
Alcohol is not good enough to remove the carbolfuchsin dye from non-acid fast bacteria because carbolfuchsin isn't very soluble in it. So using alcohol instead of acid alcohol will not decolorize properly and more non-acid fast bacteria will appear red. Hope this helps!

question:
i was working on an unknown bacteria and i was doing this procedure to identify my organism. I had a positive control on one end of the slide and my unknown on the other end; when i performed this procedure my control positive also came out blue! why!? my unknown bacteria also came out blue; so the control was compromised and i had to start over.

Not necessarily. You would need to actually do a Gram stain. Non acid fast bacteria can be Gram positive or Gram negative. The acid fast stain just shows that non acid fast bacteria don't have mycolic acids, but it cannot assign their Gram type.

what are the reasons for false positive result?
Several things can result in a false-positive, including cross-contamination between samples in the lab, contamination of diagnostic equipment, and improper use of the staining protocol, like not treating the smear with decolorizer like enough.

What result do we get if we stain the acid fast bacteria with Grams staining??
They are technically gram-positive, but I don't know that they stain well with the Gram stain because of the mycolic acids in their cell wall.

Why used acid fast staining for actinomycetes
To distinguish it from Nocardia. Actinomyces and Nocardia can be difficult to distinguish without staining. Acid-fast Nocardia can be distinguished from from non-acid-fast Actinomyces with an acid fast stain.
