    Deon Murray Some species of bacteria (but not all) go into an endospore state when conditions for growth become really unfavorable and, yes, high temperatures are one such condition.

    I'd like to know if heating the bacteria would put it in an endosporic state?

      No... Endospores form only when bacteria are in very harsh conditions... Just like lack of nutrition..(This stage is a survival strategy)
      Heating just kill the bacteria..
      Hope you understand...

    I love your videos, but I think this one should be using Malachite Green and Safranin to identify endospores. The one you are describing here is the Acid-Fast stain. Or am I wrong?

      I go over that at 4:30. That’s the Schaefer-Fulton method.

    Biology Professor Sorry I meant when it is heat fixed to the slide.. wouldn't that put all the bacteria in an endosporic state?

    I'm a little confused as to how this is different from the acid-fast stain. Doesn't that also use carbolfuscin and methylene blue?

      Yes, the Moeller endospore stain uses the same stains as the acid-fast stain. There is a longer "steaming" step between dyes than in acid-fast, but they are pretty similar! en.wikipedia.org/wiki/Moeller_stain

    Do bacteria decompose? Why are we able to keep slides for so long without them breaking down?

      Yes, bacteria decompose. Microscope slides last a long time because the bacteria are “fixed” or “preserved” in some way.

    Hi I'm a high school student and I'm trying to do this experiment (endospore staing) and i got problems buying malachite green soultion. So the point is do u know where to buy it or how to make it? And one more thing do u know the difference between malachite green and malachite green oxalate??? Fast answers would really help me thanks !!!!!!

    Your videos are great and easy to understand. However, you have a mistake in this video. In the Schaeffer-Fulton method, you do not decolorize with acid alcohol. I would adjust your video to reflect this, as the way it is described sounds like both methods require the destain.
