    Poly-A tag sequence is not for protein isolaton!( 8:16 ) poly A tail is not expressed into protein so you can´t used to fish your protein.

      Agreed, the hGh Poly A sequence is used because it can improve mRNA stability and boost translation efficiency. Also, to ensure the polyadenylation of the mRNA. In this case, a tag (hexahistidine tag) sequence will allow for protein purification.

    Question- when would u use prokaryotic expression vector and when would u use eukaryotic expression vector? Like in what situations is one better than the other?

      The difference is post translational modifications. Prokaryotic organisms are able to produce the same amino acid sequence as the gene would in eukarytic organisms but the prokaryot would not be able to modify the protein (i.e. glycosylation) necessary for the same functioning in both organisms.

    Just a question. So this expression vector will not need to be integrated into a chromosome via homologous recombination for it to be expressed? It can just be introduced into the nucleus of the mammalian cell?
