Bacterial Identification Tests: Motility Test (Hanging drop method)

Motility is the ability of an organism to move by itself by means of propeller-like flagella unique to bacteria.
If fresh culture of bacteria is available, microscopy is the most accurate way to determine bacterial motility and ‘Hanging drop method’ is a commonly used microscopic technique.
Hanging drop preparation is a special type of wet mount (in which a drop of medium containing the organisms is placed on a microscope slide) to observe the motility of bacteria.

Very helpful. In college we use the play dough instead of cavity slide. I did this experiment 4 times and I couldn’t observe any motility. So my teacher couldn’t give me the mark. I think that’s because we fix the lens on 100x power , I will try again with 40x power. Thanx
