He wants to make a Age of Compassion and to do that, he has to one break his curse and two become a god and have a consort. Also the dude who is impaled and grabbing the whole branch thing in him is a boss. He was made insane with frenzied flame and the way to contain it was to have the spear put in him. In the fight he actually takes it out and uses it as his weapon
I didnt see anyone else clarify it but yes Miquella did water a sapling with his blood, it grew to a massive size which became the base for his and his sister's city Ephael and was callee the Haligtree, which was meant to be a more inclusive tree then the erdtree which condemned omens, misbegottens, and many other species across the land.

So to trully explain the lore would require at least an hour long essay but I'll do my modest best to tie it into a somewhat comprehensible package. Both were born with a curse, Malenia with eternal decay in the form of scarlet rot and Miquella with eternal youth. Now, in base game, all we knew about him was allegedly that he was the most benevolent of the demigods. 'Miquella the Kind' as he was called actually wanted to create a new Order of kindness based upon the Order of his mother. To this end he watered one of the Erdtrees with his own blood and made a city where all those shunned by the golden order could live in harmony. Unfortunately during the shattering Mogh kidnapped Miquella to be his consort to create his own dynasty. The twist in the DLC is that in typical Souls fashion Miquella is not so benevolent as it seems at first glance. There is a reason he was also called "the most fearsome empyrian of all". Miquella wields his kindness as a weapon. He posesses an ability to sway almost anyone to his side, even those who consider him an enemy and would not willfuly follow him. One of the NPC's puts it best. "Kind and radiant, He wields love to shrive clean the hearts of men... There is nothing more terryfying." Miquella's plan was to become a new God and usher in an age with no conflict, even if it goes against one's will. To this end he needs to get to the gate of divinity and to have a Lord consort. He put a spell on Mogh and manipulated him into being infatuated with Miquella so he can get to the shadow realm knowing someone will kill Mogh for this. He also sent his loyal blade, Malenia to kill Radhan, his half-brother who supposedly once made a vow to become his Lord. Once in the realm of shadow he abandoned everything, including his body, compassion, even love to become a God and used the body of dead Mogh to house the soul of Radhan so he can become his Lord (or puppet). Miquella used everyone, including us, to achieve his goal. We use his mount the whole game. It is us who slay Mogh so he can use his body. We are the ones to kill Radhan and finish the job Malenia couldn't do. It is us that follows him into the Shadow realm where the divine gate lies. One thing he couldn't predict however, is that his power, while great, has it's limits. His charm eventually breaks and we, an aspiring lord of a different Order, whichever it may be, refuse to bend the knee.
The part where he says he's twinless is because Miquella used to have two people in one body a girl of eternal slumber saint Trina who also had Miquella's kindness so when he abandoned her he also lost his kindness which is why he's no longer a saint due to abandoning saint Trina which is also why Jt calls Miquella's age of compassion a curse because he can no longer bring a age of compassion without kindness so his age is a lie or a curse.

As for the lore questions miquella’s plan is basically to become a god and create a kind and gentle world but he’s gonna do this even if it means using his charming powers to control every being in it to force that gentleness. As for the splintered and miss her lines they refer to two things. One is him missing his sister as you guessed but the other is that Miquella like his parents is two beings in one. His other half is a woman named St.Trina. However on his path to godhood he abandoned everything including his love body and fears and even his other half cutting her out of him and abandoning her basically to die in a chasm. This is why the line “no more a saint am I” is in there.
