    You have realty done it this time Dr. Colon! This video is the best overall summary we have seen on Human Bio Science covering Lifespan and Healthspan as directed to ordinally people. Please put it into a book and embellished it with your insights on stem cells, diet. and maybe a brief discussion on Rapamycin. It would be a great book I am sure. You do a wonderful job explaining the status of this exciting leading edge science. Congratulations.

    While I take some of this already, I find already for myself that my energy level has come up. As of now I am 52 years. I don't feel like bad now. I'm also losing weight which is good. I used to weigh 502 pounds. I'm now in the 360 range, so I've come down quite a bit.

    The research on NAD has generally showed that NAD supplementation has minimum effect on aging. One person who has pushed NAD is David Sinclair but he is a bit of a cowboy. So I would certainly not take NAD boosting supplements. They are more or less useless. Another thing should be mentioned it is not necessary to target all the Hallmarks of Aging. Targeting one or two will affect the other hallmarks of aging, because they are interconnected

    NAD doesn't prevent aging. All it does is provide energy for the cells to use. Cells function better and you feel younger because of the energy. As soon as you stop taking it, you feel exactly how you felt before. You can say it temporarily reverses the depletion and lack of NAD from ageing. But the ageing is not reversed in itself.

      NAD does reverse some symptoms of aging. I got intravenous NAD+ (WAY more powerful than NAD in pill form) for a prescription drug addiction, and it literally rebuilt the exhausted receptors in my brain that were why I couldn't quit the medication.
      I noticed all these other changes too. My back pain improved, my knotted-up muscles loosened, my joint pain improved, and my guts 'purified' (wink). I am gluten intolerant and didn't know it at the time. My body also felt very uncomfortable at the beginning of the therapy and after the 14 days that were required to get clean (with no withdrawal symptoms), the discomfort was much reduced.
      So yeah, my body was purifying a bunch of crap. That crap had to have been doing a lot of damage that I wasn't aware of i.e. it was causing some symptoms of aging.
      I take NAD pills every day, and still get intravenous NAD every three or four months for maintenance, which is all you need.

      similarly, if you stop ingesting enough XYZ, you feel exactly like being without it.
      As long as NAD levels are in a good range, the aging process is much slower. why? because energy is everything on the cellular level

    I would like to add a comment to this brilliant presentation: SIRTUINS are a family of so called "Silent Regulator Genes". If they are active they express certain proteins which act as enzymes. These enzymes are also called "Sirtuins" but they are written in small letters. The genes are expressed in capital letters instead in order to distinguish wheather the subject is about genes (SIRTUINS) or their enzymatic deriatives (Sirtuins).

    I took Nuchido Time for about 2 months. . . didn't feel anything but it did make me gain a lot of weight. . . . I didn't want to get fat so I stopped taking it and lost all of that weight.

    Ok. Do you have any evidence of functional improvement after taking this product? Part of the way the NAD levels are being increase is by inhibiting it's use, which may not be net beneficial. For instance the product includes rutin and quercetin which inhibit PARP. Because of its role in DNA repair, PARP inhibition results in genomic instability and accumulation of damaged cells in cell cycle arrest. So good for treating some cancers but doesn't sound ideal to be taking on a regular basis. You highlight how interrelated all these things are so without evidence you don't know the net effect of increasing NAD at the expense of other things.
    Curious to hear your thoughts on the matter.

      Totally agree. No point in spending a lot of money on these kinds of supplements without strong evidence that they will produce the desired long-term effect on the aging process itself, rather than just affecting the level of an intermediate biomarker in the overall system.

      Current at home tests aren’t accurate as NAD is unstable and degrades quickly out of the body. Nichola talks about that in her other interviews. Hope that changes!

    I'm of the opinion that the majority of aging indicators stem from a central cause, which sets off a domino effect of other issues. Although I lack scientific expertise, it seems plausible that this could clarify why certain shorter-lived mammals, like dogs, age more rapidly than humans

    What I find weird is, exercise may activate AMPK, but actually by doing exercise, my intention is mainly to gain or maintain muscle strength, so I want to activate the growth path, so MTOR

      I think that while doing exercise you strain your body and activates your mitochondria to produce more ATP using stored energy, glucose and fats, once these are used up you are in a depleted state stimulating AMPK. That's why it's best to exercise in a fasted state. After you eat protein your body switches to growth and repair. Makes sense?

      Actually, the two of them works synergistically together. Together, they are more than sum of their parts. I have experience with them as they fixed my neuropathy after chemotherapy. That was 23 years ago.

    250 mg of niacinamide in Nuchido will likely raise your homocysteine a fair amount. Homocysteine, a hallmark of aging, raises with age and is linked to increased mortality. So does Nuchido cause you to age faster?

      This is easily addressed by taking TMG, which provides a methyl donor, alongside. This should be done with niacin, NMM, NR..or any NAD booster.

      @@Livelovelaugh684 I don't think it's easily addressed because your raise in homocysteine will only likely only be offset by TMG to a small degree. Of course, this is all testable. There appear to be many people taking NMN, NR, or Niacin alongside TMG with Homocysteine levels over 10. I don't think it's an easily solved problem.

      Methods: Thirty adult subjects were randomly divided into three groups of equal size, and orally received purified water (C group), nicotinic acid (300 mg, NA group) or nicotinamide (300 mg, NM group).
      Results: The level of 3-h plasma nicotinamide, N1-methylnicotinamide, homocysteine, the urinary excretion of N1-methyl-2-pyridone-5-carboxamide and pulse pressure (PP) in the NM group was 221%, 3972%, 61%, 1728% and 21.2% higher than that of the control group

      Just 300mg of Niacinamide caused a 61% increase in homocysteine compared to controls in a study. Nuchido has 250mg of Niacinamide. There is no way TMG is going to offset a more than 50% increase in homocysteine. You would be lucky to get a 10% offset. In all likelihood, Nuchido worsens a hallmark of aging.

      Just like Niacin will raise NAD+homocysteine. This idea to megadose minerals and vitamins is a very juvenile understanding of the body. But ofcourse commercial forces love this idea. Make 100 forms of B3 to patent and sell it for 100x price. First, test your biomarkers. Why take something you're not even deficient in. Makes no sense to me. Also get this idea out of your head, that if something is good for me, more is better! Everything is about balance and homeostasis. Look for foods high in B3. Diet is always the best way. Also there are many more ways to raise your NAD levels without side effects like exercise and fasting.

    One active good ingredient increase more by another good ingredient but at some point a god mix of people or substance can do way more than alone and in the right activity, like fasting plus good supplements can increase the benefits of fasting even way more.... so a mix of antioxidants and fasting and powerful minerals like zinc, citrate magnesium, resveratrol and omega 3 and the benefits of fasting increase by 10x?? only yoga do that increase cuality of sleep that is so important.

Look for a reputable company that does third party testing on the purity of their supplement and one that contains olive or avocado oil. Also, look at the source. Is it derived from plants or animals. From my research, animal based is the better option.
