I think Ashura Doji was the strongest during Oden time, but I don't think he grown much in strength during the last years, he was already close to his peak of power during that time.
However, Denjiro was very young and full of potential, he had to become the head of the Yakuza and build himself has a loyal servent of Orochi, also his Vivre card Databook says that he has the greatest swordsmanship skills among the Akazaya.

There is no Smile Fruit effect on Gifters because they ate a successful fruit, so those three were actually laughng. It only happens to those who ate a failed one, they don't gain the animal parts, they are just stuck with permanently smiling forever.

Big Mom riding Kaidô as a dragon could be a reference to Dr Slump or to Dragon Ball (Big Moms VA was voicing Arale in Dr Slump).
Also let's be real, if BM wanted to ride Kaidô she'd have done so in her prime already, when they were on the same ship.
