    I'm so happy I found your channel. I had been talking about growing mushrooms but never tried it. In the summer my grandma gave me a pink oyster kit just to try out how fun it was. My 6 year old son and I loved it so much that I have been trying to learn how to grow my own. I wish I had watched your cloning videos before we ate them all lol. Thank you so much for all you share about this amazing hobby (for me anyway). We really love watching. I've learned more from your videos than the many hours I spent reading prior to finding you. I want to try growing my own mushrooms and had previously come across the liquid cultures which is what led me to this video. Just a quick question... You flamed the needle instead of using the alcohol spray. Is the spray not enough to sterilize the needle or is flaming it because you don't want alcohol to contaminate the culture as you're injecting it into the grain?

    I hope you are still able to read all these comments, because I could use some help! At 12:40, you are discussing why you don’t prefer using LC commercially, and mention the high contam rate once the GS hits the high nutrients in the master’s mix. This has been my experience consistently after dozens of attempts, with a variety of manure loving LCs (I’ve had better luck with hard wood lovers.) I was only pressure sterilizing for 90 minutes, though, and wonder if that could be why? I’ve pressure canned food for decades and have never gone over 90 minutes at 15 psi, so I figured that was good, but maybe not? Or should I try a less nutrient rich substrate mix, perhaps? Thoughts much appreciated!

      Agar to Agar. Meaning that I would cut a piece of agar from a petri dish, and use that to inoculate a new plate.

    Awesome! What grain are you using? Unrelated question, do you have a cooler to store product until delivery day? We were out knocking on kitchens yesterday, picked up 3 new restaurants, woohoo.

      could it be oats? it would be nice to know some millet spawn preparation tips, I like to use millet for master inoculated by LC and then g2g to rye. I soak millet for 6h and then sterilize, but colonization of millet master jar is very slow compared to rye :(

    Good video man I still say that Paul Stamets fermented mycelium culture technique is the most effect transfer tek I have ever studied. True liquid culture manufacturing requires expensive lab equipment that most of us can’t afford. As long as you develop a good scientific method for sterilization has down grain to grain transfers are the pinnacle of mushroom growing for any style of grow

    So is the whole point of not using liquid culture for production block that if there is a contamination, you just inoculated a bunch of blocks with a contamination?

    Out of curiosity, why do you put a needle on it? Can you not just squirt it from the syringe directly?

    • @antmanmax1
      @antmanmax1 5 лет назад +4

      Contaminants. Unless you're in a clean room (and even then, because even in clean rooms shit happens), good lab procedure is to sterilize the syringe between bags, jars, etc. There's spores and bacteria floating around everywhere, and using it without a metal syringe makes it harder to sterilize (you could spray the nozzle with alcohol, but then you're risking getting some alcohol in the culture, which is no good either.)
      It's better to spend a minute now practicing good lab procedures than risking the hours you spent working on spawn that goes bad.

      It seems like if you’re not using the needle adding a needle only adds potential contaminants?

      Go through extensive lab procedures, only to not wear a face mask and talk to an audience on camera. Haha. I love this guy though he's awesome, but the mouth is probably the dirtiest part of a person besides the colon and anus. Lots of fungi and bacteria in the mouth. Also ppl are so afraid of their cultures ruining. On grain it makes sense cause that's more ideal for bacteria and mold then mushrooms. I'm fruiting oyster mushrooms in a bag I didn't sterile, on cardboard I didn't sterilize, cloned from stem buds I obtained none sterile. Also had it culture in a none sterile room with holes poked in it none sterile needle and no filter patch. Some mushrooms are tougher than we give them credit. We treat them like they're biologically weak, but think of the places they grow in nature. They're not weak, they just need a good head start and the right growing materials and conditions. We make them weak by constantly sheltering them from life.

    Germ x advanced has isopropyl alcohol, not ethanol. More than likely most hand sanitizer are going to have isopropyl or n -propanol rather than ethanol. All to say, there is no point in avoiding using isopropyl as a skin disinfectant, hand sanitizer is the same thing, especially germ x brand. Probably be better off getting 5 gallons from somewhere that does wholesale deals. I get a gallon of 99% for 12$ and cut it to 70%, and that last a while. Ps don't grow mushrooms yet currently produce tissue cultures of plants and animals , but do have lots of lab experience about to go to grad school for genetics. Starting to get very interested in mycology. Not sure how well-advanced genetics fits in to this field I've read a lot of studies on mushroom genetics but they seem to lack the technical difficulties I require to stay interested. Might just be a hobby to do some home genetic modifications or somatic hybridization.
