so MIC is the well of 250ug that you wrote in the board that is no bacteria growth but we diluted antibiotic by 1:1 with the 100ul inoculum+100ul antibiotic solution so you get MIC 125ug is that correct?
Concentration will be the same in the well...
Is there any software for calculating MIC from readings getting from spectrophotometer. In some research paper, mean of triplicate and SD was reported as MIC. Please explain the same
Yes for scan MIC method, we can use software to measure the color intensity and the diameter of pellet formed by bacterial growth...
Thank you, Mam, for your nice lecture. I have a question about the broth dilution method. Here you put 100ul media like TSB or LB in each well. Then you added 100ul of 1000ul/ml concentrated antibiotic to the first well and serially diluted it. So, my question is: when you added antibiotics to the well, which already contains media, the conc. of antibiotic will be diluted. It will be 500 ul/ml, because 100 ul media+100 ul Antibiotic will be mixed and the final conc. will be reduced by 50%. But, you didn't say anything in the video about it.
Again, when you serially diluted and added 100ul bacterial suspension. The conc. of antibiotic will be more diluted, and it will be 250ul/ml in the first well. I think you should recheck your method. Otherwise, it will mislead people. Thank you.
Of course yes... If you add media to the first well and add antibiotic, concentration will become half of the original... You need to adjust the concentration as per the concentration required according to your protocol.... After serial dilution of antibiotic you need to add inoculum and both should be in 1:1 ratio... can I use that broth dilution method not the 96 well method for my plant extract study? secondly, I want to ask in that broth dilution method don't we need to serially dilute our antibiotic in MHB before adding bacterial suspension? (you have diluted antibiotics separately in sterile d.w and then added them to the tubes). Lastly, I want to ask if can I do this broth dilution method for fungal suspension also? or only for bacteria.

Prepare the highest concentration of 1 mg/ml weighing 1 mg of antibiotic powder dissolved in 1 ml of sterile WFI. From there you can dilute as per required...

When the first well contains the highest concentration of drug for suppose 1000 ug (200 Microlitres) . If you add 100 microlitres of 1000 microgram drug to another 100 microlitres of broth (1:1 dilution) it will be half right I.e 500 micrograms.. So when you serially dilute the drug, drug concentration becomes half of its original concentration

Take highest concentration as 1000 ug/ml and go with serial dilution...

Please go through the complete video... I mentioned every thing clearly.... Take 100 ul from first well and add to the second well, again 100 ul from second added to the third well. Continue serial dilution till you reach the last well and You have to discard 100 ul from last well at the end.

In the 12th column if we are adding bacteria as well as media then why there is no growth?
Of course there will be growth if you provide media allowing bacteria to grow with out antibiotic which inhibit the growth of bacteria when used at right concentration... The growth of bacteria with out antibiotic will be considered as positive control...

No, there will be media in all the wells and only the concentration of antimicrobial compound differs... We need to add inoculum and antimicrobial compound in 1:1 ratio... Before you go for serial dilution of antimicrobial compound, first you are suppose to add 100 microlitres of broth to all the wells..

But at 26.20 minute time point of this video, you said that we remove the 100 ug of broth from the well 1 and replace it with 200 ul of antibiotic solution, this is my confusion.
OK got it.. If we want to start with the highest concentration of drug for example 1000 micrograms in the first well from 1 mg/ml of stock then we can directly add 200 microlitres in the first well with out broth and go for serial dilution of drug by taking 100 microlitres from first well added to the second well..
Now your doubt is without media how the bacteria will grow right? Here after you adjust bacterial count to 0.5 mc farland, we again need to adjust the final inoculum to 5*105 cfu/ml and then we add 100 microlitres of inoculum to all the wells right... Bacteria is diluted in MH broth... We are going to add again broth that contains the required count of bacteria

At the end of last well after dilution you will discard 100 ul so in all the wells you will have only 100 ul of serially diluted antibiotic, now you will add 100 ul of inoculum (1:1 ratio) total of 200 ul per well
