    Hello everyone,
    Good morning.
    Here comes my new video and it's all about the identification of novel antimicrobial drugs. I also explained about how we can apply the techniques of Antimicrobial susceptibility testing (AST) & minimum inhibitory concentration (MIC) in research.

    Hi mam! How to determine the stock concentration of the plant extract after drying? I've used 10gm of tulsi leaves and 250 ml of ethanol.

    Thank you mum for the great lessons . But you said we will add 1:1 bacteria solution and antibacterial extract source on the wells meaning we don't need to culture the bacteria before testing?

      We need to pick bacterial colonies growing on agar medium and make a suspension of it adjusting it to the required cfu/ml

    Madam can I know if I want to make fruit extract to perform an antibacterial activity, how should I prepare the extract. I don't think that it needs to be dried right? Or just homogenizing using a blender will be enough?

      Yaa... You can prepare fruit extracts by chopping the fruit into pieces and then blend it to get the extract, concentrate it using a rota evaporator...

    But if we add 10g in 100 ml of solvent and what will be the final concentration of plant product after roto evaporation??

      you need to check the weight of extract after rota evaporation. use that extract to carry out antimicrobial or antioxidant study which ever area you are interested in. from this stock, you need to make final concentration of 1 mg/ml or 500 mg/ml using DMSO

      To check for antimicrobial activity of any test compound, select few pathogenic strains and perform MIC as described in the video... Thank you

      Dye is to be added after incubation period to check for presence or absence of microbial growth in wells each having different concentration of drug

    I have 100 mg methanol extract stock after rota evaporation ..then how to make different concentration using DMSO(1%).should I make 1% DMSO first?

      You can prepare 1mg/ml concentration by taking 1 mg of methanol extract added with 1 ml of DMSO..

      If this concentration doesn't work for antimicrobial study, then you can increase the concentration accordingly... All the best..
