Hi Sir. Thank you for the video. I have several questions after watching the video.
1. You didn't mention about the bacteria concentration during the assay. Is it necessary to have a specific concentration? Or as long as the concentration applied is the same then it's fine?
2. What is the blank you used when measuring the OD? Is it the broth + sample (that having the same concentration as tested sample)?
3. Isn't the MIC should be at least 80% of growth inhibition?

Yes the conc. Will be in different volume. You might take 100ul, 200 ul or 0.5ml, 1.0ml like this. You cannot count bacteria to know its concentration so volume based concentration you might take. Blank is just the media what you use. There's no fixed rule for MIC it varies from bacteria to bacteria plant to plants.
Is it compulsory to get straight line in graph in MIC..???

I have a stock solution of 100mg/ml. If I take 100ul from this solution and add it to the well containing 100ul broth and serial dilute it what will be the concentration (mg/ml) in the first well. Please reply soon sir.

Is serial dilution necessary to check MIC by macrobroth dilution method? or is it okay to add extracts/oils of varying concentrations in each test tube since essential oils aren't all soluble in DMSO. plus the concentrations cant be taken in mg since not using any powdered extracts. Oils only
Yes, you can add it to the broth itself directly and keep in shaker so that if any antimicrobial compounds are there, they will inhibit the bacterial growth. Or you can follow the agar plate method, make wells and add your extract in it. Both is acceptable.
In this case you might represent your concentration in terms of ul/ml only

What solvent can we use for preparing different concentrations of nanoparticles? Can we use the broth as solvent?
Broth has the media components that will alter the media and favor/limit microbial growth. It is better to use any solvent like water if soluble or organic solvent to dissolve. Incase if ur using any solvents like methanol, chloroform etc then you can take just this solvent as one of your control too

In our normal media itself we have NaCl. So it should be fine . You can modify the normal media based on their growth. Refer research papers for more information
I used different extract of leaves then solidify and dissolved in DMSO and make different concentration for agar well diffusion method but for MIC solidify extract is dissolved in DMSO or water please clear the confusion and also DMSO and ethanol used as a control or not if ethanolic extract is used
Water and dmso both you can use. If ur using dmso take dmso as control. If it's ethanol extract, when redissolving use ethanol only and take the same as control

I use ethanol, acetone, ethyl acetate, petroleum ether extract and then solidify and dissolved in DMSO then I use DMSO as a control or ethanol or other solvent as a control

You will just add methanol to the media and the bacterial inoculum

Oils are not soluble in water generally so use a appropriate solvent which is soluble in oil. Check papers for the solvent and concentrations
