      Uhm... I tried reading a paper on acid synthesis once, and I'm quite confident in stating that, if you're not an expert chemist already, you're not even going to understand what you're looking at, let alone be able to reproduce it.

    Hot tip: when doing freeze recrystallization, put the hot container in an extremely well insulated vessel like a thermos or styrofoam box, and put that in the freezer. The slower you cool, the bigger your crystals.

      Very nice tip ! And i can add - put the hot container in an extremely well insulated vessel like a thermos or styrofoam box, and do not put that in the freezer, let it cool slowly to room temperature, then put in fridge and let it cool to fridge temperature, and then put that in freezer. Then you get largest and purest crystals. And use minimum amount of solvent for recrystallization.

    1:55 - Just a _slight_ correction. Although many classical psychedelics are agonists of the 5HT2A receptor, it's actually the 5HT2A-mGluR2 complex, an oligomerized receptor complex consisting of both the 5HT2A receptor and the mGluR2 (metabotropic glutamate receptor 2) receptor that is responsible for psychedelic effects. There are drugs out there like Lisuride which acts as a partial agonist of 5HT2A, but doesn't result in psychedelic effects because it only binds to the uncomplexed 5HT2A receptor. The uncomplexed 5HT2A receptor has a similar, yet distinct, binding pocket when compared to the complexed 5HT2A receptor in the 5HT2A-mGluR2 complex. So, a drug that binds to 5HT2A doesn't necessarily mean that it's a psychedelic drug, but there's a good chance it will be.

      @@sciencething1449 Yeah, this whole "it's not the 5HT2A receptor, it's actually a 5HT2A receptor complex" thing confused the fuck out of me when I first heard it. Then, I went to a lecture on receptor dimers at university and it started to make sense. Basically, GPCR's (G-protein coupled receptors), can join with other receptors to make a dimerized receptor i.e. a combination of 2 or more receptors. They can have similar effects to each receptor on it's own, but they can also have effects that are completely different.
      It's an emerging field of research. It's so big, and we currently know very little. Like, if activating the 5HT2A-mGluR2 complex can result in psychedelic effects, who knows what effects messing with other dimers can result in?

      @@sciencething1449 I think that the mechanism of how/why GPCR's dimerize is currently unknown for certain, but it's a hot topic of research. Expert opinions range from "omg these particular GPCR's never dimerize" to "Of course all GPCR's function as dimers."
      What we do know is that the way that these GPCR's dimerize is through linking of certain domains (extracellular domain, transmembrane domain, c-terminal tail) from each GPCR through disulfide bonding, and non-covalent bonding. This also explains how a dimerized receptor can have different pharmacology. If the binding pocket is located on the extracellular domain in each GPCR, and the receptor dimer involves two extracellular domains linking together, that could change the shape of the binding pocket and mean different ligands can affect the dimerized receptor compared to the undimerized receptor.
      The GABA-B receptor is quite interesting in this regard, because it can only function as a heterodimer of GABA-B1 and GABA-B2 receptors, and the mechanism of how this occurs is known. GABA-B1/B2 have minimal function on their own, and they need to dimerize to make the final GABA-B receptor, and there's a very particular mechanism to it's production.
      GABA-B1 and GABA-B2 are encoded by two separate genes (the genes are called "GABBR1" and "GABBR2"). When the GABA-B1 mRNA is produced from the gene, the mRNA then enters the endoplasmic reticulum, where it is converted to protein, the GABA-B1 receptor, and inserts itself into the membrane of the endoplasmic reticulum.
      But on the GABA-B1 receptor, there's a particular protein sequence which causes it to be retained in the endoplasmic reticulum until a finished GABA-B2 receptor comes along, which causes a conformational change in both receptors. This leads to both receptors subunits linking together through the c-terminus tails, producing the completed GABA-B receptor. The conformational change in the receptor also allows amyloid precursor protein (APP) to bind to it, which causes the finished GABA-B receptor to form a vesicle, releasing the completed GABA-B dimer from the endoplasmic reticulum, and be shuttled to the cell membrane.
      Since dimerization is necessary for the function of GABA-B, this could just be a way of ensuring both receptors dimerize into a functional receptor. Maybe other receptors can dimerize just by being close together on the cell membrane. Maybe there are other mechanisms of dimerization with other receptors. Unfortunately, we don't know for certain.

    Salvinorin A is such an underrated compound. I'm willing to bet that the majority of people who dislike Salvia did so because they did little to no research on the substance. They probably expected a euphoric high similar to cannabis, and I can't even blame them. Due to dumb drug laws, Salvia was marketed as an incense not for human consumption which meant they couldn't list the correct dosages, causing people to fill a whole bowl of the stuff when all they really needed was a small pinch.
    I think the best way to use Salvia is as a tool to explore consciousness while letting go of all expectations. The moment you expect an outcome is the moment the drug turns on you.

      I'm so glad that when I got into Salvia, there was already a dosing guide on the dmt nexus. Using a scale with a sane level salvia extract (~10x) was a game changer for me. The other game changer was discovering that salvia in a changa mix was amazing. A guaranteed ticket to carnival/circus land.

    Salvia has a bad reputation but I think it's because most people had no idea what they were getting into. It's effects are powerful and peculiar.

      I would love to try salvinorin A extract because I hate the smell and tasteof the leaves so much. I have a feeling the trip would be less dysphoric if I didn't have to experience that taste.

    Salvia was by far at the top of my favorite trip list. I always explain it as a 5 minute LSD trip, it honestly feels like hours, last time I ended up in a snow bank. Make sure to have a good trip sitter, a lot can happen in 5-10 mins.

      Once had some 5x extract and it made my skin crawl but not much else. Then I won some 60x extract and thought "why not?". Whoosh. I thought I was 6 years old again on my way to school on a snowy day. I could feel the icy cold metal of a lamppost (in reality my glass containing water device). It felt so real. No trace of current reality was left. Then, after an unknown amount of time I was back and totally freaked out by what happened. Not a bad experience but enough to make me NEVER want to try it again.

    Recommendation: flash chromatography column packed with lab grade clean cotton, clean sand, charcoal. Boom. Edit: check out this book: Techniques in Organic Chemistry: Miniscale, Standard Taper Microscale, and Williamson Microscale This is what I used for ochem back in my undergrad program. It's a solid manual on techniques and is cheap now.

    Hello dear friend
    My main field is electronics technician and graphic design
    But due to my interest in chemistry, I am also active in this field
    I saw that you used devices and funnels for filtration several times
    I don't think there is a need for so much complexity
    A 20 ml syringe
    Remove the piston
    Squeeze a piece of cloud or Kleenex tissue
    Push the tissue into the syringe until it reaches the outlet
    Press it with a tool that is not sharp
    Now pour activated carbon up to 1 cm
    And now dissolve the crystal in water
    and pour into the syringe
    Put the piston in place and press gently
    and over
    I used it many times in the extraction of ephedrine and so on
    The result was excellent

    If you have access to a -80°C freezer or some dry ice it helps tremendously with the wax/chlorophyll issue, extract is light lime instead of dark green. Also just realized it was like 15 years ago that I last did this extraction, damn, time does fly...

    I feel you so mutch with the aktivated charcole issue, I had the exact same problem with an extraction I did, only difference: I had a Hand crank centrifuge... yeah my arms were unuseable for an houre or so after turning the the centrifuge with my pure muscle power... again and again...

      ​@@chemdelic yeah if you can get around all the Wax and fats they have and are able to make a pure product that would double your sub count over night lol only time I was able to get a clean product was to use dry acetone and let it soak over night then filter the top clean part it made a brown crystal that looked amazing but sadly every extract I've made or bought isn't as strong as taking the hole powder itself 😔 but finding a way to make it easier on the stomach is my goal because it sits so heavy in my stomach it makes it hard to eat and you just feel like shit when you drink it it's so bitter

    another way you could have done that decanting was by using a funnel with a small size filter fret plugged with diatomaceous earth. another way would be old fashioned filter paper with a slight vacuum pump 2 micron will clean it up nicely.

    You may want to clarify at the end that the mg scale a lot of people use off amazon isn't good enough. You need like a $10,000 dollar scale to measure safely for sub miligram measurements and it needs to be properly calibrated.

      yeah a safer way to dose it would be to measure out like 50 mg with a cheap scale, dissolve that in some quantity of ethanol, and then dose it volumetrically onto blotter paper or some kind of plant matter for smoking. That way if your scale is a little bit off it won't make a big difference.

    Is the removal of the chlorophyll in rhe latter stages just for aesthetic reasons, or does it mess with the efficacy of the product?

      I’m not chemist, but if you don’t get it out, it seems unlikely to crystallize properly. Regardless I’ve performed extractions, and getting chlorophyll out is something you want, it could interfere with the steps after it.

      You can smoke the leaves directly and you would have effects from the Salvinorin A. It mainly is an isolation of the psychoactive compound. It just changes the style of consumption as leaves are less concentrated and this is WAY more concentrated. Though mine mainly was for aesthetic reasons. Plus it is much easier to dose when you have the exact isolation of a pure compound rather than an unknown concentration with impurities!

      Probably don’t want to inhale the vaporized chlorophyll. On the other hand the chance of introducing a more toxic impurity through the extraction seems high for the general caliber of chemists doing this extraction. Most of the videos I’ve seen the product looks more like dried leaves than extract.

    Did it once....My main trip was awesome. My job in life was to lay on my back in a meadow while stamping out multi-coloured 'crazy paving' slabs out of the gooey stream that was flowing overhead....
    However , the off-ramp was so horrifying that I can't bring myself to try it again...I believed I'd being held captive and sedated for 6 months and I 'realised' that the sedation was wearing off giving me a chance to escape. I somehow managed to get up, stagger down some stairs and out into the street. I felt like I was moving through electrified gel.

If they want to come lock me in a cage then they will, I will not censor, hide, or change anything about myself to appease some pseudo overlords. I am hurting no one and I am proud of my actions, I will always do them in the light.
      To your second point yeah I've done it a handful of times and you're 100% right in that it's intense, words fail to describe what is experienced on the deemz. Last time I did it a voice told me to stop for a while, you'll know when it's time to come back, and its time to come back.
      Im a lab tech and a coworker did the extraction for me last time but I think I need to remove the training wheels

    Like filtering every pull straight on. Look what you pour out of the bottle and how green it is, after the night. That is caused by all the stuff, you did not filter out straight.
    Pull time of 3 times one minute, is 2 times and 57 seconds to long. It's plant material. Just grow/buy 20% more of it, to cover the losses.
    Get Salvia leafs, not powder. Powder is only good for a vitamin rich smoothie.

    Nice work. There are simpler methods, but given how potent a substance this is, I don’t want to spell out Salvinorin A Extraction for Dummies here. Hint: the easiest, but not most productive, methods I have tried are basically just like recrystallization, but starting with leaf powder. The change in solvent temperature and volume does most of the work. Extract the leaf powder into a solvent that you would use as a solvent for recrystallizing salvinorin A (ethanol works well). Maybe do two 24-hour macerations. Filter the combined extracts, then boil down to concentrate (how much to concentrate here is important, but you’ll have to experiment). Put the concentrate in the fridge for a day or two. Then filter off the crystals. They can then be washed in boiling hexanes. Then in boiling distilled water. They should be quite clean at this point, but you can purify further by recrystallizing in ethanol.

    I am so sensitive to Sally D, that I tend trip HARD on small doses of dried leaf extract. It definitely changed my way of thinking after a few very deep spiritual trips. When you say you will see God, I can back that 100% I can't imagine where pure Salvia would put me. It's my 3rd favorite psychedelic and for me, comes nowhere near the same as shrooms or LSD. Salvia is on it's own plateau. It's get the most respect from me.
