  • If you’ve been following Dr David Sinclair, you know he updates his supplement protocol from time to time.
    And in each interview, he gives one piece of explanation to what he does.
    And certain supplements that appear in one interview, he does not mention them in other interviews.
    Sometimes it’s hard to follow his protocol today.
    Since, I know Sinclair’s work since 2007, before he went public in 2019.
    Here, I have collected for you from a variety of sources, including Sinclair’s book, trying to figure out EVERYTHING he takes today and how much from each supplement.

      @@henrikmadsen2176 TMG consists of glycine with three attached methyl groups. It’s naturally produced in the body, found in certain foods like beets, and available as a supplement. It’s involved in a process known as methylation and can help lower levels of homocysteine - which improves cardiovascular health and protects against dementia. It’s a very safe and well studied supplement that also may increase athletic performance and improve insulin resistance.

      @@roba.8907 Thankyou for your great answer. I have seen Dr. Sinclair advise taking Glycine and NAC together (GylNAC). I assume for that I can then use both ordinary Glycine or TMG? I think I have seen Dr. Sinclair recommending taking NMN with TMG, because of the tri-methyl gruops), but now NMN is banned by FDA, I assume it's not sold any longer, and maybe also not safe to take? So is Glycine is cheaper than TMG, I might just go with the Glycine-NAC combo.

    Read an article awhile ago that man who had calcification of heart valve and was due to have a valve replacement. He started to take higher doses of K2-7 and asked for tests after taking this high dose of K2-7 and his valve showed significant decrease in calcification. He opted out of replacement valve surgery and continued to take Mk2-7

    I would really like to clarify about Quercetin - it’s not safe to take it every day, it can damage healthy celks, and I dont think Sinclair is doing it everyday either….the study that he brings up (and I can look it up) used quercetin in hit-and-run fashion meaning 3 days in a row then 6 weeks break. Because there is no point to keep taking it everyday - once u remove senescent cells.

      Yes, he said in his podcast - recorded in late 2021- every day. Maybe he has changed that since. I'm with you on this question and expand on that in the resveratrol research series which includes touching on Quercetin. Currently 2 videos are out of many in this series of research :)

      From Dr Kirkland, the “father” of senolytics research: “Since new senescent cells take 10 days to 6 weeks to form, these senolytic agents can be administered in a hit and run manner - once or twice if there is an insult like radiation that’s not repeated, and do that periodically as new cells form. In those experimental situations the drugs can be given once every two weeks or once in a month and that has the same effects as giving these agents continuously. We are not trying to occupy a receptor or inhibit an enzyme, not looking for a steady state of the compound in blood, we’re trying to get rid of a cell type, allow these cells to kill themselves.”

      There was also something about the role of the senescent cells in wound healing and other processes, so there can be downside in their complete removal. Let me look it up, it was an interesting argument and reasoning behind not taking quercetin and other senolytics continuously but in that pulsating manner

      Quercetin doesn't kill senescence cell alone. It is necessary to use together with other drug to have this effect.

      I currently take 375mg of quercetin with 500 mg resveratrol with olive oil 5x a week. Not sure if its good or not. I am eating way more vegetables now than before so maybe I don't really need to have it as often. :/

    Interesting- If I remember correctly, apples have fisetin and quercetin - pretty much the same amount as in most senolytic activators - I guess the old adage is true- an apple a day keeps the doctor away😊

    I used to take 20 100 MG K2 MK7 (liposomal pills - 2 gm all at roughly the same time....) for 21 days to reduce calcification in the arteries. It made me feel better and I followed up with more normal doses. Also used boron (9 mg), fish oil, and full multivitamins/multiminerals - it all reduced arthritic stiffness as well. High doses of magnesium and potassium also were helpful, and so were liposomal pills. Whenever I take significant doses of calcium, I try to take Vitamin K2 (complex - both MK2 and MK7).
    NMN (liposmal - 1 gm) or Niagen (300 mg) should be taken with quercetin (500 MG) and TIMG (1000 MG) for better results.

    Thank you for this concise account.
    I take 1gm NMN daily sub lingually.
    I have ordered 100 gm resveratrol and Fisetin on line and am in a quandary about dosage.
    I take Vitamin D 10,000 and Vitamin K2 90 micrograms daily.
    At 81 I want to stay on track.
    I read Lifespan and Super Human and have used NMN since 2018.
    I purchased a NMN Resveratrol mix on line for my son which he takes sublingually. This was because one supplement was easier for him to remember.
    My GP prescribed Metformin 1gm when I asked to keep my Blood sugar under control and it coincidentally reduces my blood pressure.
    Give my son 250gm Metformin. Advised him to ask his GP for a script. He has high BP and my father had diabetes so reasonable request. He
    My GP said he has been taking Metformin for years as preventative health measures. His father too had diabetes. It is a cheap drug and very safe.

      @@WellnessMessiah I found I was using 2mg which is overkill. Also taking 5gm Creatine Monohydrate for muscle strength and balance. For older women it helps the bladder to stay functional. I put it in my fizzy multi vitamin and Vitamin C (5gm) drink. There is no substitute for ageing well.

    Amazon is banning sales of NMN products in its store because of an FDA letter stating that NMN is not considered a dietary supplement as of November 2023. This is a reversal of FDA's previous decision. Their stated reason is to protect a pharmaceutical drug development application for a patented version of NMN.

    He did mot mention apigenin to block CD38. He could have added R alphalipoic acid to quercetin to boost NAMPT. TMG good. All three are required to assure the salvage pathway for NAD+. He vould have added NAC when combined with glycine in TMG creates glutathione, the master antioxidant. There's so much more to know.

      I agree wholeheartedly! And, I honestly don't think he looks all that young for age 52. Especially considering the fact that he's been taking Resveratrol daily for nearly 20 years!!

    I'm a little surprised David takes so much of so many of these compounds. Most of them are synergistic to the point of actually amplifying the effects of one another.
    The effects of Metformin and Aspirin, taken together, is more than the sum of each taken alone.
    The effects of Metformin and Resveratrol, taken together, is more than the sum of each taken alone.
    Metformin, Aspirin, Resveratrol and Quercetin are all synergistic, with many overlapping effects.
    I have taken all of these, and in similar doses (but 1 g Metformin per day) but only for 4 weeks, as a purge. I wouldn't want to risk doing this long-term.
    The only one I take every day is Metformin and that's because it's the only one of these compounds that has been proven to extend human life and has an excellent safety profile. On the other hand, one study on rats showed that Quercetin increased the occurrence of tumours.
    Starting Monday, I'll be taking the polyphenol, Pterostilbene together with NR as they seem to have a powerful anti-cancer effect when taken together. Pterostibene has 4X the bioavailability of Resveratrol so I'll only be taking 100 mg per day. I'll also be starting with sulforaphane and eating much more broccoli together with mustard seeds (trying looking that one up).
    When I was taking Resveratrol and Quercetin (my purge) I mixed them together with soya lecithin, on a slice of wholemeal bread. Both the lecithin and the wholemeal bread boost absorption, especially the lecithin, which has a strong affinty with water. I also mixed in 500 mg of VitC which I had in paste form, in capsules. The Vit C and lecithin help to protect the Quercetin, the lecithin in the stomach and the Vit C in the bloodstream, so it has more time in your system before being metabolised.
    Anyway, thanks for keeping us up to date.... really appreciate your videos!

      I just had a look on Pubmed and all the papers I can find suggest Quercetin tends to have anti-cancer effects, both preventative and also therapeutic. I'm not saying there are no papers to the contrary (couldn't find your rat one!) but I would go rather with the weight of the evidence if I were taking the supplement. That doesn't mean the studies I found were any good of course.

      Really sorry about your mother. There is no words for this kind of loss. I am a bit lost at the moment on K2 since I saw a video of Brad Stanfield with desapointing results of a K2 study. I keep on taking it but it was unsettling.

    I do not have a prescription for metformin. So I decided to substitute it with Berberine since it is very effective in reducing blood sugar. I would like your opinion on this….

      Hi, Berberine has a wide array of impacts. It's more complicated - from my research - than metformin (3-pronged effect, metformin just 1).
      But one of its mechanisms indeed overlaps with metformin.
      Personally, I do not take it at the moment and have more personal experience with metformin.
      I can tell you that I've seen a massive reduction in cholesterol with my clients, so I suggest tracking that too.
      Dr. Peter Attia is very smart on Berberine and uses it in his clinic, so I'd go to check his recommendation. The brand here matters and I know Attia is using Thorne research, albeit it quite expensive.

    Great video! Just one thing: Taking drugs like Metformin is not natural and not healthy, it has side effects. If you do not eat sugar, you do not need Metformin! I do low carb - high fat - medium protein and IF and most days OMAD. Do sports in a fasted state. Take Pterostilbene on my resting days, not when doing sports.

      Hi Kristi, I would add the original molecule from Metformin is actually created by plants, not humans. They used the plant which has had some nausea effects, for many decades. It seems that any useful drug comes from plants after all. It is a drug, as you said, today.

      @@WellnessMessiah Hi Rimon! Yes most molecules in medications come from plants, mushrooms, bacteria or animals. That does not make them harmless. Metformin has bad side effects. Not eating sugar and carbs has no side effects and is the most natural way to get healthy. And is not expensive.

    I saw a video by Dr. Berg where he stated that you can take 10,000 IU's of vitamin D3 with 100mg of vitamin K2 (MK-7 form only due to it's increased bioavailability) and not be concerned with calcium build up in the arteries, therefor you might be safe doing that.

    After having watched all your videos I'm still insecure about how much resveratrol to take (30-50mg or 500mg or even 1g like Sinclair does??)....Hopefully your forthcoming videos will help finding the right dosage...
    As far as i know wheatgerms contain much more spermidine than the sources you mentioned.

    Hi, thanks for the great info. Do you know the anwers to the following questions? 1. do you have to keep NMN refrigerated 2. how could I take NMN when I travel (refrigeration not possible) 3. how can I see if NMN is still good 4. when I get NMN delivered it is not refrigerated, how can I make sure it is still good 5. is NMN really harmful once it changes due to not being refrigerated?

    Just started resveratrol (for 1st time) at night after a meal, with coffee with the 2 tsp of olive oil and coconut milk. Not as bad as thought it might be. (after watching one of Sinclair's videos, ad lib.) Have to wait and see literally, results. Read about coffee as antioxidant. Also have read that it stops vitamin absorption. (Hopefully just slows time of absorption.)

      Dr Sinclair is a fraud. Every video he is in is filtered to make him look younger. But unfiltered video shows every bit his age. From the receded hair line to the crow's feet and deep nasallabial folds. For someone who has studied aging for decades it hasn't worked. Now that his company got NMN banned, even though it's a substance the body produces, because he wants to profit off of it. I wouldn't listen to anything the guy says. His resveratrol controversy is enough to shed light on his scam...but then to get NMN banned? Right.

      Taking both has a synergistic effect, and correct me if I’m wrong but I think I remember seeing that fasting can cause some muscle loss with athletes and other highly physically active people which may be undesirable for some

    Thanks for the overview. You mentioned coconut yoghurt a couple of times. He states sever times that he is vegan and uses plantbased (coconut) yoghurt. Also I think he said in one of his podcast he takes 5000ui vit d3 a day.

      Dr. Sinclair is a scientist/experimenter. At a point in time he'll tell you what he's taking but that can change anytime because sometimes he carries out these experiments on himself.

    I have enough different kinds of supplements to start a health food store. After seeing your presentation on resveratrol I stopped. It appears to me that in high doses used continuously it might be dangerous. I'll stick to red wine and berries.

I had my long overdue annual physical Monday and my lab reports look good. Triglycerides are a little high. A1C was 6.3. She gave me no advice on supplements I haven't taken except to say don't. Very disappointed. However nobody knows everything. Maybe she's too busy working to follow research on all of these supplements. I'm trying maqui berry extract to see what it does to my blood sugar.

    How about this study concerning A-HgM (Metformin f.ex.) exposure in patients with T2D was associated with reduced risk of MS in patients younger than 45 whereas in patients older than 45, exposure to A-HgM was associated with an increased risk of newly diagnosed MS, particularly in women. Would this then also apply to Berberine?

    Why doesn’t Dr. Sinclair have made the supplement and and market it and letting others to do that and make business out of that? As far as I know pioneer in this reverse aging, is Sinclair. But I don’t see any supplement/product has come out of him other than free guidance, while others make business out of it. Which is not fair in my opinion.

      I speculate smart consumers could get NMN from other countries if it is banned. I hope the governments wouldn't care so much about importations for an un-risky supplement such as NMN.

      The book goes into detail about why antioxidants don't reverse aging like science thought they would 10 or 15 years ago. The reserveratrol is working to reverse aging using a different mechanism, so you can't just replace it with an antioxidant

    I just ran across this channel today (Feb. 6, 2023) and it seems pretty good. I'm a fan of Dr. David Sinclair and Dr. Brad Stanfield, and I've attended two high-quality international conferences on longevity. Almost all this stuff is quackery and snake oil, but perhaps Rimon's content is not.

    This is another very informative video. I just recently found your channel and really enjoy the content, especially when you dive into the details of Sinclair's comments and the studies.
    Since this one is about supplements, have you considered doing some videos on liposomal supplements? A lot of these longevity substances have very poor bioavailability. Using a liposomal delivery system dramatically increases the amount that gets into the bloodstream (almost all of it from what I've heard) and also facilitates entry into the cells where it is needed. I'm in the process of switching to liposomal (or phytosomes, which is very similar and does pretty much the same thing as liposomes) supplements where they are available for the stuff I take.
    One last thing, Metformin is mentioned in this video, and that is not typically available to people. You've also mentioned Rapamycin on your channel I believe as a longevity molecule. It turns out there are natural mimetics of those substances. On that reasoning I added ashwagandha, EGCG, and ginseng to my stack. See the study below for scientific support:

      There is no doubt that liposomes are a superior delivery method.
      However, from my research, the company has to make them right and it makes it more expensive than most people can afford - including myself at the moment.
      But yes, there is no doubt, that if there is an effective liposome supplement, it will increase absorption pretty significantly.
      The cheapest way to do so, so I heard, is to mix the supplement in a mixer with something like sunflower lecithin, hoping it will form a liposome.

      Hello, I would be interested as well on your look over some liposomal supplements. I looked on Internet to make it myself (mixer, ice, emulsifier) nonetheless I don't know how important is the size of the liposome in the delivery method. Homemade liposome might be pretty big.? Home blender might not have enough shear force? Further emulsifier might not be very healthy as some of them might weaken the gut barrier ? If the supplement is in a liposome, maybe it is not helping oher parts of the body that would need it as well? Anyway lots of questions 😊

      the studies were carried out with non-liposomal active substances. If these substances were to be replaced by liposomal substances, which are much better absorbed, the dose would probably have to be greatly reduced. The only question is how much the dose should be reduced. A manufacturer indicates a 6-fold better absorption of its liposomal NMN (250mg thus correspond to 1500mg). Can this be true?

    When talking about vitamin K2 supplement dosage it’s important to be more specific on the type. I believe he was talking about MK-7 K2. You also have MK-4 which in most supplements is dosed higher than MK-7.

    Do you know Vitamin K2 and Vitamin D3 are fat soluble. So if you do not have a good Liver and Gallbladder. You can not properly breakdown and hence cannot absorb these vitamins properly meaning you do not know how much of Your daily intakes

      But for many hours, heat increases oxidation. It can turn trans resveratrol into cis resveratrol, which doesn't work at all.
      But immediately, again, - seems fine to me!

    I am aware of the resveratrol you are taking which is a water soluble molecule I believe. What do you think about Pteristilbene which appears to look like resveratrol but is more lipid soluble? Would taking both the water soluble & lipid soluble form be more beneficial in your opinion? And does pteristillbene work under the same or similar mechanism as resveratrol?

    May I add to the Spermidine question that you find HIGH concentrations of it in wheat germ. It is up to three times the concentration of mushrooms. At least that is what the web says.

      I add wheatgerm to my tinned tomatoes in my regular salad. Tastes ok. So pleased I can now eat less of it I was trying to get close to 1gram which isn't really possible. Great source of vegetable protein too

    Do you address, or can you address, Berberine as an alternative to Metformin?
    My doctor is not really up on longevity research, and she refused to prescribe Metformin to me. However, Berberine is touted as a natural alternative to Metformin, with many of the same benefits, plus others. Of course, I purchase Berberine online, so it's more accessible.

      Hi Christopher, Berberine has a wide array of impacts. It's more complicated - from my research - than metformin.
      But one of its mechanisms indeed overlaps with metformin.
      Personally, I do not take it at the moment and have more personal experience with metformin.
      I can tell you that I've seen a massive reduction in cholesterol with my clients, so I suggest tracking that too.
      Dr. Peter Attia is very smart on Berberine and uses it in his clinic, so I'd go to check his recommendation. The brand here matters and I know Attia is using Thorne research, albeit it quite expensive.

    So am I to understand just to be clear that he takes virtually all of these supplements at the same time or around the same time in the morning? I don't hear him or yourself mentioning that he takes anything later on in the afternoon or evening. (I heard him mention some people take NMN at night time). Just curious, is he taking D, K2, Resveratrol, Quercetin, Fisetin, NMN, Spermedine, and Tri methyl glycine all in the morning?

      Hi Ryan, I can only know what he says. he know Vitamin k2 needs fat, so I think he takes with meal in the evening. He is doing intermittent fasting and skp breakfast.
      Fiseting and quercetin, NMN, RES, spermidine, he takes in the morning.
      TMG - I mentioned he stopped mentioning this. Metformin, he say in dec 2021 he takes at night.

      My understanding is that berberine is very effective at flattening a glucose response in the body, but you also want to use it very sparingly. i.e. only take it before a carb heavy meal, such as the rare time you go for pizza and ice cream.
      I would not recommend using it in a routine manner.

      @Lynn Russell Fantastic that your friend reversed type 2 diabetes, but I would primarily attribute it to meat and veges diet not the berberine 3x a day. Cutting out processed food, refined sugars and sticking to whole foods such as meat and vegetables is all that is required, berberine is on the margin.

    The poor man’s metaformin you can get over the counter (Amazon) as Berberine, which he used to take. IF you’re a weightlifter (you all should be), take it every other day or just on days you take off.
