Vander has been turned into Warwick through the experiments of Singed. The LoL model for Warwick is definitely more “wolf” like and I think they chose to make the Arcane version more human in the face to tug at our heart strings.
isha is a girl, in episode 3 very quickly when isha jumps on jinx with the gun and VI gets off of them, she tells cait its a little girl. The whole jinx/isha part of act 2 is my favorite.

Warwick is a LoL character, yes. I think Singed too. (but I hardly checked out LoL, it's too toxic, especially for beginners)
