I'm pretty sure it doesn't count towards completion percentage, but there's achievements tied to it. With that out of the way, iif you missed some enemies in the crossroads before it became infected, there should still be uninfected versions of them around. I dunno where off the top of my head but a google search would get you there pretty quickly if you wanted to do it.

You need Isma's Tear to swim in acid, I think I get it around the 30 minute mark on episode 7 of this playthrough if you need a visual
