      Cuvette need not be full. It should be at that level where light will be able to pass completely. So even if the cuvette is only filled half or more than half and still it will give results.

    Sir suppose NADH was at 0.1 but NAD+ was at 0.2 then what should we do ?

      NAD+ doesn't have any epsilon value so overall 0x 0.2x 0.5 cm would have been zero. But let us say NAD+ have some amount of epsilon value and also concentration is different that is 0.2 mM. Then first take absorbance of NADH separately and then absorbance of NAD+ and simply add them as absorbance is additive in nature.
