Wayne Knight is proof that big guys are funnier. This scene is hilarious largely because of the physical element of the comedy. Big guys are always best at physical comedy. Chris Farley is another great example. Just look at how Newman is sweating profusely. It's not the same with skinny guys

Chris Farley IS the example! He had a huge heart and actually could act. Look at Tommy Boy and Black Sheep again and watch the parts where he generally acts. His physical comedy was really unmatched! Man can still make me laugh. His Oberdose was an unfair hand he was dealt.

It's hard to pick out a single "funniest" scene from Seinfeld. But this one would always be a top contender. 2 total idiots concoct an over elaborate scenario to get out of paying a simple traffic citation. And then it falls apart due to their inability to stick to the plan, along with their frequent argumentativeness and bickering.
