I had a test today, and we had to draw the lines in the test tubes, I vaguely remembered what the drawing of the experiment from the revision book looked like and I got it right?? I’ve rly impressed myself today
Here's a step-by-step explanation of the Meselson-Stahl experiment: 1. **Labeling DNA with Heavy Nitrogen (15N):** - E. coli bacteria were grown in a medium containing a heavy isotope of nitrogen, specifically nitrogen-15 (15N). - As a result, the DNA synthesized by the bacteria during this period incorporated the heavy nitrogen into its structure. 2. **Transfer to Light Nitrogen (14N) Medium:** - After several generations of bacterial growth in the presence of heavy nitrogen, the researchers transferred the bacteria to a medium containing the more common, lighter isotope of nitrogen, nitrogen-14 (14N). 3. **DNA Sampling at Different Time Points:** - The researchers took DNA samples from the bacteria at different time points after the switch to the light nitrogen medium. 4. **Centrifugation and Density Gradient Analysis:** - The DNA samples were then subjected to a process called density gradient centrifugation. This technique separates molecules based on their density. - The DNA samples were spun in a centrifuge through a density gradient, which allowed the separation of DNA molecules based on their nitrogen content. 5. **Observation of DNA Bands:** - After centrifugation, the DNA formed bands at different positions in the density gradient tube. - If replication followed the conservative model, the bands would show distinct heavy and light DNA molecules. - If replication followed the semi-conservative model, the bands would show an intermediate position, indicating DNA molecules with a mix of heavy and light nitrogen. 6. **Results Confirming Semi-Conservative Replication:** - The results of the experiment showed an intermediate band after one round of DNA replication, supporting the semi-conservative model. - In subsequent generations, the intermediate band persisted, and new bands corresponding to fully light DNA appeared, confirming the continuous incorporation of light nitrogen into newly synthesized DNA strands. The Meselson-Stahl experiment provided compelling evidence in favor of the semi-conservative model of DNA replication, as opposed to the conservative or dispersive models.
The Ecoli cultured in N15 was transferred into the Ecoli cultured in N14 And in the first generation we get to see the hybrid N14N15 and later in the second generation we get to see 2 density gradient band one is the hybrid N 14 and 15 and other is the N14

I'm learning so much but feel like I'm at da club
I'm a German student and I understood the experiment only with this video in a foreign language 😅Well done!👍

For easy teacher..
I'm from Pakistan🤗😂
I loved this video; simple, precise and entertaining(the music in the background). since it has one beat all through it isn't distracting
The accent tho 😂
Great vid btw. Really cleared it up for me 👍🏻
even i was thinking the same, i mean he went from American- British to idk we can't call that indian either.
This was so helpful! You explained this in a much more precise way than my textbook! Thank you so much!
Short nd sweet.. Good one 👍..I subscribed expecting even more good videos
Thank you so much for this video ma’am. It really helped me to understand the concept
Here's a step-by-step explanation of the Meselson-Stahl experiment:
1. **Labeling DNA with Heavy Nitrogen (15N):**
- E. coli bacteria were grown in a medium containing a heavy isotope of nitrogen, specifically nitrogen-15 (15N).
- As a result, the DNA synthesized by the bacteria during this period incorporated the heavy nitrogen into its structure.
2. **Transfer to Light Nitrogen (14N) Medium:**
- After several generations of bacterial growth in the presence of heavy nitrogen, the researchers transferred the bacteria to a medium containing the more common, lighter isotope of nitrogen, nitrogen-14 (14N).
3. **DNA Sampling at Different Time Points:**
- The researchers took DNA samples from the bacteria at different time points after the switch to the light nitrogen medium.
4. **Centrifugation and Density Gradient Analysis:**
- The DNA samples were then subjected to a process called density gradient centrifugation. This technique separates molecules based on their density.
- The DNA samples were spun in a centrifuge through a density gradient, which allowed the separation of DNA molecules based on their nitrogen content.
5. **Observation of DNA Bands:**
- After centrifugation, the DNA formed bands at different positions in the density gradient tube.
- If replication followed the conservative model, the bands would show distinct heavy and light DNA molecules.
- If replication followed the semi-conservative model, the bands would show an intermediate position, indicating DNA molecules with a mix of heavy and light nitrogen.
6. **Results Confirming Semi-Conservative Replication:**
- The results of the experiment showed an intermediate band after one round of DNA replication, supporting the semi-conservative model.

What an amazing explanation 👍👍🤗
Thanks from India🇮🇳🇮🇳
Very thoroughly explained, liked it!
Very easy to understand
If some N14/N14 can also be made in the second medium, why can 2 N15s not find each other here similarly?
