How heavy is this? I can't imagine holding a leather briefcase on one hand all day, it would make my shoulders get tired. This is if I put some stuff in it, about 600g of water and toiletries

Hiya :)
The bag weighs about 1.5kg on its own. It does get a bit heavy with laptop/water/accessories in there and I certainly wouldn't want to be using the handles at that point. The shoulder strap is very comfortable though and takes the weight off your arm.

Does it fit a 16" laptop and a water bottle?

Hiya!
A 16" laptop won't fit in the specific laptop section. It will fit in the main compartment though.
A bottle will fit alongside the 16" laptop but it depends on the size of the bottle, the slim metal bottle most people use would fit fine, as would the standard plastic bottle you get from a shop. You won't get a 750ml/1l bottle in there, or any of those larger "motivational" bottles.
