Hey Gary, I just wanted to chime in 1 more time to let you know about this experiment i did with peptones.. i added peptone to the water when i was boiling my oats.. after i bagged the grains up and inoculated them , the 3lb bags are almost fully colonized after 5 days...much faster than normal. Cheers bro

Have you considered doing this experiment with each isolate in triplicate/quadruplicate? You could do it on a smaller scale so it's not wasteful, but it would help a bit with any variability that may be unrelated to the isolate biology so that you draw the correct conclusion of which isolate is best.

What would the expected genetic outcome be from spores harvested from a specific phenotype? For example, would M3 spores produce only M3, or all phenotypes?

just make sure you label as you go and before you switch strains and pay close attention - this was a huge issue in my previous jobs because of the stakes of lab testing so it was engrained in me hah

2 weeks from spore to clean culture, 2 weeks on grain, 2 weeks incubation, 5 days pinning, 3 days first flush, 19 days second flush (1 1b and .6lbish for flushes)
Hello I am a soon to be Uni student but I cant decide on what field I want to go into, microbiology/ biochem has caught my eye and I want to give some basic mycology a go. May I ask what injection filters are for? Do they have in this case the same use as micropore tape, to regulate air intake aseptically? I Cant find a conclusive answer. Since I have not that large of a budget and since I cant find a pressure cooker/pot with a pressure gauge in my country... a lot of my budget has been eaten up by that, I was wondering if injection filters and micropore tape interchangeable at the cost of chance of contamination? Are both micropore tape and injection filters necessary?
yes it filters air and helps to filter air if liquid is being displaced in a liquid culture as well
For the last one and half months i just can't make spawn without trichoderma infecting the grain. Make spawn from tissue culture only, doesn't use agar or lc. Need some advice.
it’s that time of year - this month and october is really challenging because the normal spore load is 10x or more - my advice is up your sterility standards (mop the floors, clean the workspace, add filtration) or wait it out 👍🍄❤️

How are the slants packaged for international delivery on etsy?
we heat seal them, send them with appropriate paperwork in a bubble sealed thermal envelope
Were those bags still warm when you inoculated them? Did you mix the 2 types of pellets before inoculation or did you figure you could mix it at the time of inoculation?
Weird how it can differ from flush to flush. My first, had a bunch with cleft caps from growing in tin pans and as the rust and mycelium color got worse, the second flush came out nicely. lol!
they were about 75ish degrees and I didn’t premix the pellets only after inoculation

yes it helps during all phases to keep things clean

@@FreshfromtheFarmFungi I wonder if it would tend to be relatively fibrous. Of course, proving it out as a cultivar (vs an anomaly) might be more of a labor of mush-love than a profit-making endeavor.
