Consider the following:
Luigi just went through hell to get Mario back. This was one single night in which he faced all sorts of specters, ghosts and maybe even some demons in a mansion Mario went ahead to check.
This is the first time Luigi is ever established as a scaredy cat, yes, and given how he reacts to every single major encounter with a ghost of some kind, any other person with a similar mindset would've died from cardiac arrest.
After several fights, he fights and succesfully defeats King Boo possesing a Bowser puppet, again, all of this to get Mario back.
This man went, faced all the trials a spiteful oversized Boo threw at him, succeeded, returned to E. Gadd, delivered all ghosts, had to be comforted for thinking Mario was dead after all his trials, and then, after all of that, he could finally see his brother again in the flesh.
This was his first major adventure, and it was hell. Obviously, his mind was shattered by the end of it all and seeing Mario's head is stuck on a vent frame in a rather comical way was all he got to cope with everything that had happened.
He laughs (and cries -- he starts crying first, mind you), not because Mario has his head stuck in a vent frame and is all wobbly and confused, but because this hell is finally over and he is finally reunited with his brother

The most touching parts are when Luigi and Mario reunite and hug. True brotherly bond at its finest, even when Mario stands on Luigi for a solid 15 or so seconds. And Mario calling Luigi #1 shows his respect towards him as his equal as well.
