How Does It Work? | AAV Based Gene Therapies

In the mid-1960s a small and unusual type of virus, known as AAV, was discovered. AAV cannot replicate on its own and it causes no known diseases in humans.
These unusual properties have enabled us to use AAV as a vector for ground-breaking gene therapies for patients. Let’s take a closer look at how they work.
For some people, a defective gene causes a life changing disease, but the AAV vector can deliver a therapeutic gene directly to their cells to alleviate the effects of their defective gene.
The virus introduces genetic material into the cell nucleus, where it forms an episome that does not integrate into the cells existing DNA. It also dilutes in dividing cells, thus preventing transfer to new generations.
The transcription of this gene can help to reduce or eliminate symptoms the patient experiences from their defective gene.
AAV technology is still growing, with clinical trials underway for a range of targets that include genetic disorders, neurodegeneration and HIV infections.
