Betty Ross' repressed anger and the negative self-image she has due to the way her father treated her (as Samson says to Gen. Ross in the movie, "In all the time we've been together, Betty never talked about her father -- now I understand why.") expressed as a transformation into The Harpy (a later transformation after she had made peace with her father resulted in the Red She-Hulk, much like her father's transformation into Red Hulk). Dr. Leonard Samson decides to experiment on himself to see what effect gamma rays would have on a "well-adjusted" mind, and it taps into his childhood fascination with the Biblical hero Samson and the pulp hero Doc Savage, hence Doc Samson -- basically an idealized version of himself. A man who feels less smart than his peers and has low self-esteem becomes the hyper-genius egomaniac called The Leader. A man who is beaten as a child manifests an alter ego who is angry and child-like who needs to be "the strongest one there is," so no one can hurt him again: The Hulk. A man with a childhood obsession with the biblical Samson and a deep resentment of being pushed around by bolder, more physical men becomes Doc Samson, a Doc Savage style brilliant gentleman adventurer. A repressed, timid, short lawyer becomes a bold, aggressive, 7-foot She-Hulk. And so forth.

He gets exposed to gamma rays from a device that was intended to draw radiation off the Hulk in conjunction with psychological stimulus to cure Banner's transformations. That obviously hasn't happened yet at this point in their interactions: he's only just now meeting Banner.
