Only one, capture antibody will be coated to the plate, later antigen will be added, later detection antibody is added. The antigen forms sandwich between capture antibody and detection antibody. So only one antigen.

But why are we using two antibodies, when we can work with only one antibody and link it with the enzyme. And when there was direct and indirect ELISA was there so what is the need for this one?

Sandwich elisa is used to detect antigens where as indirect is used for detection of antibodies

Sandwich ELISA generally has higher sensitivity and specificity than the direct ELISA because two antibodies are involved in capture and detection. Since antigen does not need to be purified prior to measurement, sandwich ELISA is a better choice for the analysis of complex samples than the direct ELISA
