    This video is the last video in a series on Genetic Testing for Mental Health. It discusses the COMT (Catechol o methyltransferase) gene. This genetic SNP will be of any interest to anyone that has an interest in genetic testing for mental illness, genetic testing nutrition, genetic testing for autism and genetic testing for anxiety. Genetic weaknesses in the COMT gene are relevant to depression, anxiety, bipolar disorder and panic disorder.
    In this video Dr Janelle discusses symptoms associated with the COMT gene mutation, and she explains why changes in the COMT gene lead to these symptoms. Finally diet, lifestyle and supplement changes you can make to help support your COMT enzyme activity are discussed.

      Hello Dr., I have my COMT gene with the polymorphism Val158Met. You have mentioned as the "best" scenario, but actually it's not, at least for me, because I was diagnosed on the bipolar espectrum, and it's exactly like this: at some moments I'm very anxious, neurotic and stressed, but soon I feel depressed, apathetic and with lack of motivation. It's hard to find a balance, I'm on medication now, but I intend to discover more natural ways to achieve this balance. Greetings from Brazil.

    I think more people should understand this. Im 46 and have struggled my whole life with anxiety problems, and then starting in my 20's insomnia problems. I burned out over and over again because of work and life events. I feel lucky to be alive actually with what I've been through.
    I got the Methylation test done recently and I have the COMT mutation. So it explains my struggles.

    So interesting! I figured my heterozygous COMT was a bad thing but it sounds like it's the best COMT configuration. Although I'm not homozygous COMT, I could really relate to that slide: workaholic tendencies, thinker, activity in the morning, stress management, calming activities at night. I can't tolerate caffeine at all or stimulants in general. Last year, a doctor mixed epinephrine with the lidocaine he injected in me and I ended up in the ER. Of course, the ER doctor did nothing and acted like I was crazy. I'm excited to watch your video on caffeine. Thank you for putting all this info out!

    I love the bucket analogy, because I cannot remember all the processes as I read Ben Lynch‘s book. This will help me explain to my daughter what I think is happening in her brain. She certainly won’t read the book because she does not have the ability to maintain focus. Thank you so much for this brilliant video.

    Interesting! My partner took my methylated b vits and they made him really jittery, plus he can't handle caffeine. Makes me think he may have the double mutation!

    I have homogenous mutant COMT V158M and COMT H68H genes. I definitely can't deal with caffeine- Matcha is the worst. I end up shakey Anderson scatter brained as my thoughts are running through my head too fast to follow anything through properly. Haven't found any B12 impact. I can definitely be an over worked stress'o'holic at times! Will have to try the Phosphatidylserine! Thanks for the tip😊

    My first video. Very good.thank you. I subscribed. I have my genes on25 pages, but not everything is perfectly explained, and updates are always occurring. Thank you for a very practical view on the gene (summary) and then the food/ supplements/ behavioral traits that correlate. I appreciate it.

    I have the G/G genotype indicating fast COMT however I am severely prone to anxiety, insomnia, high energy, high motivation, and high intelligence. So perhaps there is more to the equation than COMT alone.
    Also, I read some advice that consuming more methyl groups will actually be beneficial for people with fast COMT activity since your body depletes them more quickly. Quick depletion means you need to quickly refill.
    So far I’ve tried supplementing with betaine, inositol, and I’ve increased my choline intake significantly from consuming 6 to 7 egg yolks per day, I’ve also added methylated B9 and B12.
    I am feeling a lot better than I did before starting these supplements.

      Magnesium supplements are high priority for anxiety. I'd also rule out zinc and iron deficiency.

    This makes so much sense! Thank you for the explanation! My naturopath has put me on a fairly high dose of methyl b vitamins to help my adrenals and my anxiety has shot through the roof! I’ve also never coped well with caffeine or dark chocolate. Just wondering - would you recommend avoiding ginseng as it is a stimulant too?

      I have slow COMT and avoid methylated vitamins. There's one called B-Minus that is all the Bs except B12. They're all unmethylated
      You can choose which kind of B12 is best for you. I tried a normal B12 and it made me feel manic like I was going crazy or something. Very scary. If you want to add a methyl donor of some kind be careful and start slow. I just take Creatine since it frees up the body from making it's own creatine, which takes up a lot of methyl donors.

      Just found out I have this slow COMPT and mfthr! Thought I was loosing my mind over the years because of the anger anxiety and panic atacks. My mood was all over the place and sleep was getting worse each year. Glad I took the gene test! Good info @DrJanellesinclair Best of luck to everyone battling these problems! Stay away from benzodiazepines unless you have go take them. Been battling benzodiazepines withdrawals for 4 years and it was the scariest thing ever. I had panic atacks / anxiety. Wish I would have taken gene test or technology had allowed me to take gene long ago. It feels like a loosing battle but hang in there people you can change quickly and feel amazing if you eat right take your supplements exercise and change bad habits Also stay away from stress best you can. I have only been on supplements for slow COMPT and mfthr a few months and feeling like a superhuman especially with slow COMPT. 💪✌❤

    But it isn't that simple that low dopamine are corresponding to ADHD. Low COMT activity actually increases chances for having ADHD significantly. It's beacase too high dopamine in prefrontal cortex can lead to overload. Too high noradrenaline causes restlessness.
    Indicators of low COMT activity caused ADHD are co-problems with anxiety, psychotic signs and high ability to hyperfocus.

    Great video! Thank you. Do you recommend taking Sam E for a slow comt if have both missing? Not expressing too many of these symptoms, but susceptible to stress. I’ve heard some people getting worse with Sam e.

      @@DrJanelleSinclair I agree....I took SAM e several years ago and it drove my blood pressure higher. I think I was taking 400 mg. a day which I realize now was a high dose.

    Was just diagnosed with COMT Met/Met and MTHFR, I’m really having a hard time understanding it all and this gave me a better idea. I’d love more information. I have debilitating symptoms, cyclic vomiting, I basically can’t function. My anxiety is unbearable

    I was re-watching this and noticed the methy-B12 part. That's exactly when I had an AH-HA moment, because I had been taking methylB12 and it was just doing something really weird to me, like I was ramped up and haven't been able to come down.

      It’s so crazy because during chemo I had to take folic acid to counteract some of the poisonous aspects of the chemo drug and my temper was crazy.

    I have one copy of MTHFR C677T allele + heterozygous A1298C + Slow COMT! Should I avoid dark leafy greens because of their high phenol content or should I consume them because of high folic acid ? what about Keto diet do you think I should consider it to loose weight or is it going to be harsh on my liver?

    🙋‍♀️ Question-I’m COMPT G/A-does that mean I don’t have problems with neurotransmitter balance? Since the intermediate COMT wasn’t really discussed it left me wondering. 🤔
    My M.D told me to cut out/ limit intake of berry’s and green tea and avoid L-Tyrosine & high protein meats which actually seems to help me most of the time. So confused. 🤷‍♀️

    So if I have MTHFR and fast COMT, would I do better with methyl groups from methyl b12, methyfolate, TMG, etc? I’m assuming I break down methyl groups quickly and would therefore require more methyl groups?

    I'm a homozygous AA mutant.
    I've spent my whole life wondering what the hell is wrong with me. Turns out, nothing, but my insatiable need to know really helped when I finally did have something wrong with me.
    I don't ever sleep, never have. I have PMDD, and small inconveniences can feel like the end of the world.

      Omg! I know how you feel I’m the same I have 2 C677T and recently I’ve come to think I have the COMT snps because supplementing made me worse.     Interestingly, SNPedia actually suggests ADHD is associated with A;A or Met/Met (worrier). This is obviously a bit strange given ADHD is linked to low dopamine levels and A;A individuals tend to have a lot more of it in their prefrontal cortex. Here you listed it as being connected to G;G. To me, it makes more sense that it would be. However, I am A;A and do have ADHD. Of course this could be entirely unrelated but I always wondered if there was some connection.
    I also have an MTHFR mutation so I have taken methylated folate over the years to help with this. So it was interesting to hear it might harm me in other ways I wasn't considering.

      Afaik ADHD can actually be cause by too low OR too high dopa. Yeah mthfr can cause your dopa to be low, then when you're methylated you start to get normal dopa levels which becomes too high for you to handle because of comt.

    I must have the double +/+. Inget easily bored. I like moving to new places and I get so much pleasure from being in new places and doing something different. I am not scared of changes and risks.
    But I hate not being able to tolerate caffeine and too much stress

    Great video thank you. Curious however as to your thoughts on diet. Is it particularly important to lower protein and increase carbs to lower dopamine? What about fats, especially saturated fat? I have the ++ slow comt gene and unfortunately cannot eat many carbs either due to oxalate sensitivities.

      Protein is very important for neurotransmitters. Don't reduce it in your diet because they also help create other balancing neurotransmitters. I don't think most people eat enough protein.

    I see most of the traits of the slow COMT variant in myself. I have also experienced extremely prolonged effect of certain types of medication, especially psychiatric. Any sleep aid will turn me into a zombie for days despite me taking half of what other people normally do. Could this also be a sign that I have the slow variant?

      I have comt slow. I'm a mess with agitation, restless,"anxiety" but nothing is wrong. It dictates my life& work choices, sucks. it's about half the day this goes on. Anyway I also take 1/8 of a melatonin& I can sleep 16hr. FYI

    I can't afford to have this testing done. Doctors don't do it, and in Texas naturopaths aren't covered by insurance.

      Thanks contrarymary. ☺️ I haven't come across any data saying it's contraindicated. Just start on a small amount and increase slowly if you are concerned.

    For you dopamine you can also supplement with copper, L tyrosine is converted in to LDOPA through (b6, iron and folate), LDOPA breaks with magnesium and b6, Dopamine breaks in to noradrenaline by Vitamine C and COPPER, (Hole Food because of the co factor tyrosinase, just vit C íll lower you ceruloplasmin levels by not bean about to connect cooper to ceruloplasmin and also drops copper levels, best is acerola pulp) Noradreline breaks by SAMe in to adrenaline.
    Check your copper levls and ceruloplasmin fisrt.

    What if the other COMT SNP: rs5031015 is GG but COMT SNP: RS4680 is AA. What does that mean? Is that balanced?

      rs5031015 (G/G):
      • Higher COMT expression: The G/G genotype at rs5031015 is associated with higher expression of the COMT enzyme. This means the body likely produces more COMT enzyme, which can somewhat counterbalance the slower activity from the Met/Met variant of rs4680.
      • Potential effect: While the rs4680 (Met/Met) variant slows enzyme activity, the increased expression due to rs5031015 (G/G) means the body is producing more COMT enzyme. As a result, one might experience a slightly faster rate of dopamine breakdown than someone with both the low-activity rs4680 (A/A) and low-expression variants of rs5031015.

    I have slow COMT tested and it fits 100%. Now, how do I reduce phenols in food? They are in everything that's healthy, like vegetables, fruits etc... Any recommendations would be greatly apprecciated! :)

    My COMT testing came up as high estrogen, and I have high dopamine, was encouraged to take I3C. I have had anxiety and bad depression. I didn’t realize magnesium and vit C were helpful. My b12 seemed fine but B6 low.

      Remember to stay off caffeine too. It'll be one of the most important things to follow. Maybe some phosphatidylserine if you're not going to sleep easily

      I am COMT ++ and use Indole3carbinol ...have used it for over 10 years and will always continue to use it. It was recommended & a requirement by my Alternative Medicine MD .when I started using bioidentical hormone therapy....it helps to clear excess estrogen from the body. I take 200 mg. with breakfast or 100 mg. at breakfast and 100 mg. at lunch.

      It's heterozygous. You're not slow or fast, you could say it's in the middle. It's the best result.

    I have two COMPT mutations. rs4680 GA (+/-) and rs4633 CT (+/-) And lately, I have been experiencing extreme anxiety and having daily anxiety attacks. I also have three MTHFR gene mutations 😑 so I get it so hard.... but we CANNOT take folic acid, so please don't recommend that. I'm definitely going to look into the alternatives you mentioned to the B12 though.

      She recommended folinic acid. Not the same thing as folic acid. Do your homework.

      folic acid is synthetic and therefore your body does not truly recognize it nor use it as it would if not synthetic plus it can interfere with absorbing Folate ... I actually use folate but I see Folinic Acid is also suggested if Folate is not the best fit for you

    Thank you so much for this video.
    So, I have one MTHFR mutation, and I am AG heterozygous for the Val158Met which you mentioned is the best type to be ... but I struggle with anxiety and panic attacks. I’ve done therapy, I live a healthy lifestyle etc, but five years later, I’m still dealing with it (it’s better but still affects my life) and it feels physical (physiological) rather than “I’m worrying all the time” etc. So confusing.... if I have the most ideal COMT .. why do I deal with anxiety. I also had my neurotransmitters checked and I have perfect dopamine but low serotonin, low PEA and low adrenaline / noradrenaline ... makes no sense to me!

      Brittany anxiety isn't just caused by our genetics, there's lots of other physiological causes. I suggest that you check out my videos on a) sugar and anxiety b) Zinc deficiency c) Brain fog, depression and gut health. All of these videos are very relevant to anxiety. Adrenal fatigue may be another contributing factor. Let me know if you have any other questions after you watch these videos. If anxiety runs in the family, zinc deficiency can be very important. (The videos I mentioned are videos that I created first, so look back right to the beginning)

      @@Kayyy8585 I personally think that your mthfr and comt gene variations won't play a significant contributing factor to the anxiety and depression. If you were homozygous for a mthfr SNP it might. I would investigate nutrient deficiencies, gut health or hormones.

    I had my COMT tested and it doesn’t say if I’m AA or GG. It says V158M and ➕⬇️/➕⬇️ in red. I have no idea what this really means.

      No you'll have to look at the results for the COMT gene. They are two different mutations.

    Yep AA comt and homozygous for mthfr also. So as soon as i try treat mthfr with methyls it revs up my comt worse. I've just tested my whole blood histamine and homocysteine so im very interested is seeing the results as to over or undermethylation.
