    I had a lab practice exam yesterday and this guide helped me a lot. The lab teacher had taught us to put a drop of water on the slide and then touch it with a loop containing the cell sample in its tip. The results were dubious at first, but then I remembered this method and we managed to get a very clear Catalase + result.

    Please, can you elaborate why the culture has to be 18 to 24 hours old?

      Aerobic organisms must be taken from an 18 to 24 hour old culture. Organisms lose their catalase activity with age. For slower growing anaerobic organisms, an older (24-72 hours) culture may be acceptable.
