Luffy's actor watched a decent bit to get a good feeling of the character for his audition, and after he got the role definitely watched the entire anime. Zoro's actor had been following it for years (keep in mind, he's Japanese, and One Piece is just so big in Japan there's no way you don't know what it is if you live there) Nami's actress has been a huge anime fan for years with Nami being pretty much her favorite character ever and is fully caught up on the manga as far as I know. Usopp's actor I don't specifically know but it's clear he has at least some decent knowledge about the anime. Sanji's actor took cooking and taekwandoo classes to better get in the role and also definitely has seen a solid amount of the anime for it from what I know
Sanji probably the one who have the least knowledge on one piece, Sanji and Luffy both are new fans, but inaki caught up fast.. Zoro is japanese and Nami is long term weeb and maybe Ussop too but they definitely heard alot of rumors too

No role requires that EVER. If an actor has learned everything about the role they are playing, they either did so in their own time or already knew the character prior to playing the role. The VAST majority of actors have no obligation to do any such thing, they just need to learn from and imitate the script they are given. They are absolutely NOT required to read or watch the entire thing just for some context because that is what the script is for.
