For those about to ask, the Liquid Culture Premix = 1 part by weight LME (light malt extract) and 4 parts Corn Dextrose. Andy stores it in a repurposed whey powder container, but there is no whey in the formula. And if you are doing the mentorship program, step it up and do a RUclips video for Andy as part of your hands on learning experience. I just tried the 15 ml centrifuge tubes. I feel they are too small and will be switching back to 50 ml.

@@Xalos369 Honestly, I love them all. Even the baddies. Scorpy is one of my all time favorites though, great arc from bad guy, to understanding his motivations bad guy. D'argo of course, and Rygel both are truly awesome, and Za'an was one I really loved and was sad to see her depart the show. I don't know that I could pick a favorite, except John. I really feel like that show got a close to honest reaction of an American being whisked off to an alien setting. Rarely do you get to see the snarky humor in Scifi like that.
Excellent update on making slants. Thank you Andrew, appreciate the time you put in making these RUclips videos for us. Don't feel bad about plugging yourself. You give us tremendous amounts of knowledge and when that knowledge changes you pass it on, so ya ... plug away, and honestly I like. You mentioned you can store slant cultures for years, then later mention you need to keep up with them like a year or so later. Does that mean you have to redo the slants every year or two? Would spore prints last longer? Thanks again Andrew, take care.

I purchased some pre-made slants with a popsicle stick added, but I had purchased the lid that had an air filter and injection port. I left them at room temp, and they just shriveled up over time, and even a few little mushrooms tried to grow in them. I scraped what agar culture I could off of the popsicle sticks and also flushed them with distilled water to suck up any mycelium I could get, to inject into liquid culture-to test if it was still viable, since 95% of the agar was gone (they weren't made as good as yours-yours have more in each tube--mine was an etsy purchase). I'm just wondering if I should just do solid lids, instead of an air filter patch and injection port-to make them last longer. That, and evidently a fridge. lol. Oh and I love it when you get flustered, it makes for good videos. lol
I would never use slants with filters on them. Filters fail, and the point of slants is to store for the long haul. Not too mention the drying out problem you just mentioned as well.
Andrew, I used your Oats soak method, pressure cooked it for 2 hours in half gallon jars and 1 bag. When cooled I covered the filter to prevent moisture loss. How long, at room temperature, will it still be viable for inoculation?
Andrew, please recommend an induction unit capable of dealing with a Presto 23 qt. There are a lot of options but I like experienced references. Thanks David
What is the purpose of the toothpick? When you are ready to revive, do you just add water and suck into a syringe like you would when making an LC from a dish?
You put hardwood in there because it gives you a even better chance of long term storage. If something happens with the agar, a lot of times the wood piece will still be perfectly fine, meaning the mycelium of the mushroom variety has innoculated itself in the wood, and you won't lose your culture. It's more so a extra safety and longer term storage
I've seen a few 'How to set up slants' and I've seen a few 'how to make an agar transfer from a plate', but I've not seen a 'how to make a transfer from a slant'. Maybe a subject for a short?
A lot of my tubes warped and became concave in the middle of the tube. Anyone have any idea why that happened? I followed these instructions. Maybe i should loosened the caps? I put them on hand tight, but didnt crank them, just like suggested in the video.
So i don't have to dissolve the agar end stuff in hot water? Because one time I tried to sterilize directly in tubes but the agar mix settled at the bottom, maybe it was my old PC now i have a new one.
No, I never pre dissolve my agar. I use hot water when I make the slants and we shake the tubes to mix it in a little. It dissolves when we shake it for the most part.

Andrew, what’s the Celltreat model number on those centrifuge tubes? Most I’m finding have HDPE caps which I thought melt in a PC/autoclave.
