- The next episode of The Cure Chronicles features Raif Derrazi, an HIV/AIDS advocate, vlogger, fitness model, and natural physique bodybuilder.
In 2012, Raif was diagnosed with HIV. A week later, on his birthday, he was diagnosed with AIDS. Following his diagnoses, Raif refused to let the virus control his life. He built his strength and physical fitness through bodybuilding and started sharing his experiences online in hopes of inspiring others to become their best selves, no matter the circumstances.
Today, Raif is a host on +Life Media, a multi-platform brand dedicated to bringing conversations about HIV back into the mainstream and ending HIV stigma. He has partnered with organizations like the AIDS Healthcare Foundation and NMAC in an effort to change how people view living with HIV, and is set to be an ambassador representing the US at the Gay Games in Hong Kong next year.

What is the possibility of combining AGT103-T and EBT-101 Therapies?
In combining EBT-101 and AGT103-T, the patient would first take the gene and cell therapy treatment, AGT103-T, which involves HIV-specific CD4 T-cells that are genetically modified to resist HIV, by having a sample of peripheral blood cells collected from the patient and CD4 cells that recognize HIV’s Gag protein. A harmless lentivirus vector is then used to insert genes that disable CCR5 receptors, where HIV uses to enter cells. The modified CD4 cells are then multiplied and returned into the patient’s body.
The same patient would then take the CRISPR-based one-time gene therapy, EBT-101, that will utilize AAV to deliver an intravenous infusion intended to functionally cure the HIV infection. The CRISPR cuts out or excise the HIV that has wrapped around the DNA cells. This therapy would harness the AAV at a relatively low dose to deliver the treatment, which would deploy CRISPR-Cas9 and two guide RNAs that target three sites within the HIV genome.

@@Lee-1 FDA approved American Gene Technologies (AGT) to move forward with Phase 1 Clinical Trials, in Maryland, Baltimore & Washington. D.C, for an HIV Cure Program back on August 11, 2020. On November 9, 2021, AGT announced that an independent Data Safety Monitoring Board (DSMB) that overseas the AGT103-T human clinical trials, and found no serious adverse events from treatments of a third patient.
On September 15, 2021, FDA accepted Excision BioTherapeutics to begin trials testing EBT-101.
AGT103-T is a cell editing and gene modifying product made from a person's own cells, while EBT-101 is a CRISPR gene editing treatment.
2100 is almost a century away lol. The results for both AGT103-T & EBT-101 should be released between 2027 and 2032.

Keeping medication up to date in a third world country is not so simple, and seeking the medication itself generates stigma.

Would this potential treatment be effective to cure HIV-2 as well?
I know that HIV-2 is rare in EU/US and as a consequence there's not much research in treating/curing it, but in fact some of us are living with it without any hope and unfortunately there's almost no available information regarding HIV-2 research.

In one word: yes. If you're indetectable, then you're untransmissble. Chances of contaminating somebody is almost null under this condition. So yes, HIV positive people can marry and have kids via natural means without risking their spouse's or their child's health.

So he had Aids with no idea of having HIV+, hmmm you, first of all, lose weight and due to that virus, your body will pass through a lot of illnesses etc. His story is awkward.
My story may be awkward but it is true. I also did not have much issues with weight. I have always had lots of illness my whole life so it didn’t seem too out of the ordinary that I was experiencing random ailments. None of them was bad enough to make me want to go to a hospital (having no health insurance) until my throat really was too painful.

I resonated with your story because like you, I so got used in getting sick especially in my 20s. My diagnosis is still a hard pill to swallow because I was physically fit and have been working out until I got sick in January. That time, I couldn't even look at myself in the mirror because I lost a lot of weight, something I worked hard for in years. And that time, I was also recovering from sickness and felt like the Universe just wanted me to lose. I still cry thinking that I was too close to dying hadn't I had myself get tested because of a recurring flu that didn't seem to go away anymore. I cannot say I'm mentally fine now but your channel has helped me a lot in coping with this condition. I told myself I'm gonna be like you and went straight to working out again even at home two months in my treatment. I'm still afraid I may not be able to get my body back just like how it used to be but you're there as an inspiration. I'm still at that stage of acceptance and it's truly hard but people like you are very helpful. I hope to get an undetectable result very soon too and thanks you for inspiring so many of us.
I was the same as him. I got used to a lot lf sickness but in my case, I didn't really think about it because my body looked normal. I was working out and was able to put on a lot of weight in two years of bulking, which I eventually lost due to AIDS wasting in a matter of weeks when my immune system was already too low. My weight loss was gradual but it became very drastic in January when I caught a flu. My face began shrinking and I lost a lot of fat and muscle weight too in four weeks.

I can relate was given. my result on my 21st birthday in 1992. Also went super healthy for 3 years or so but once healthy decided to say yes to everything in life big mistake but learnt. Did the AZT then cocktail got sickling story many twists and turns.

AGT103-T, in theory, could be use preventatively as a vaccine, but it would be significantly harder to make due to the lack of HIV-specific TCR enrichment that are seen in PLHIV. While AGT103-T does not deliver CCR5 delta 32 (the CCR5 mutation that naturally blocks HIV in certain individuals), AGT103-T does deliver miRNA to prevent CCR5 from expressing on the cell surface, which makes cells resistant to HIV entry. AGT103-T achieves the same functional effect (phenotype), but it is not the exact CCr5 delta 32 genotype.
AGT103-T was recently FDA approved for human clinical trials. Through clinical trials, results of the clinical trials is typically takes between 2 and 5 years (end date between 2027 and 2032) because each time a patient is infused, they are extensively monitored for side effects, efficacy, and variations in their response to AGT103-T.
The two gene therapy approaches, that are human clinical trials, in the hopes of functionally curing HIV, (1) AGT103-T, as discussed, which as we see id developed by American Gene Technologies and (2) EBT-101, which is developed by Excision BioTherapeutics, have two totally different approaches for functionally curing HIV. AGT103-T grants HIV resistance to the T cells that are responsible for hunting HIV, allowing them to do their job and fight HIV like they would for any other virus; this includes continuously fighting the viral reservoir. EBT-101 is a temporary pulse of CRISPR-associated gene expression that aims to cut out all HIV from the human genome, but since these CRISPR-associated genes cannot be continuously expressed, they need to cut out 100% of HIV proviruses during their expression window, otherwise HIV can just replicate and return.
Data from both therapeutics, after successful human clinical trials, will be needed in order to know how well both AGT103-T & EBT-101 works and most importantly, whether or not a AGT103-T + EBT-101 combination therapy would be warranted.

I have a question; In January 2022 I had a test and I had a negative HIV test result.
In March 2023 I had a positive test result and CD4 = 130 viral load = 280,000. I have been taking medication for a month and CD4 = 195 viral load 328. Is it normal that CD4 has dropped so low in a year? 😢😢
