  • Biochemical tests are very conventional & popular method for the detection and confirmation of bacteria after isolation from any sample. This video presents a detailed procedure of several common Biochemical Tests (TSI, MIU, MR-VP, Lysin, Urea, Citrate) those are needed to confirm, specially, a gram negative bacteria according to ISO 6579-1:2017.

    Hello, can I know what is the volume of media are needed in broth tube for the culture before biochemical test is performed?

      It varies on type of media. Some are 5 ml per tube, others are 10 ml for each tube

    In the step three: pure colture - 01, need to inoculate the colonies from XLD to TSA plate. Which XLD plates need to choose? Those previously obtained from selective enrichment of RVS broth or MKTTn broth, or both?

      Subculture to TSA from XLD is important. You should choose colonies all XLD plates (from MKTTN and RVS, both)

      @@MicroChemsExperimentsI would have another question. The ISO does not specify the non-selective medium to be used, can alternative media to the TSA be used? For example plate count agar or yeast extract agar?

      @@MicroChemsExperiments To confirm the presence of salmonella, all biochemical tests must be positive (in particular TSI, Urea and Lysin). Is it possible the tests give a positive result for colonies obtained from RVS selective enrichment and a negative result for colonies obtained from MKTTn selective enrichment (or the reverse)? In this case what would the outcome be?

      Culture your suspected colonies on Trypton Soya Agar, Nutrient Agar or Plate Count Agar. Then used colonies for biochemical tests

      Free fatty acids (FFA) are produced by the hydrolysis of oils and fats. Since FFA are less stable than neutral oil, they are more prone to oxidation and to turning rancid. Thus, FFA is a key feature linked with the quality and commercial value of oils and fats.

      @@prince_ranaofficial398 Selenite Broth is used for the selective enrichment of Salmonella spp.
      E. Coli and other coliforms are inhibited in this broth.
