Its a long task. Its difficult to explain here. Neverthless you can subculture your colonies on different selective & differential culture media like MSA, Cetrimide Agar, XLD, SS agar, Chromogenic Coliform Agar, MRS, MYP agar, etc. After that do biochemical tests of the selected colonies for identification.
Thank you for uploading this video.. and it's really helpful and understandable too.. please do video for detection of staphylococcus spp and vibrio spp.
Don't cover the lid of plate until medium is solidified N make sure the agar is completely dissolved in Duran bottle while heating in microwave This can be achieved when u see the medium is boiling clearly i.e bubbles formation.
hi your video is very resourceful but can we use selenite broth instead of RVS and MKTTn broth? and also another question, what is the name of this method used? thank you
No. Peptone water is used for Non-selective enrichment. Non-selective enrichment can not be achieved by distilled water. So you must use Peptone Water.
1. Advantage: This method involves double enrichment steps which increases detection possibility and facilitates to detect Salmonella more selectively, thus this is the more accurate method compared with other methods. 2. Disadvantage: MKTTN media, frequently, becomes not available in local market and there is no replacement media of it.
Why does solid foods need 24hrs for incubation after homogenized with buffered peptone water???..Not only for salmonella detection but also for enumeration of TPC??
Wauu. This is really a good video. Can anyone share with me a full document where I can find medium types used to enumerate bacteria founds in food. Please help if possible. Have a nice weekend
Tomorrow i wll face auditor in micro lab its very helpful to me thnkyo so muchh
All the bestttt

Very on point and interesting video.
Much appreciated
Thank u so much mam ...its very informative
You are welcome
Please, give us the link for part 2 and do the video for bacillus cereus
Excellent explanation thanks 💐👍
Thank you
We have to take pre-incubated plates pf XLD or BSA?
Good job, I've got a cultured plate with distinct colonies and I'll need your help in identifying the bacterium and medium used. Thank you

Please make videos for v.c. v.p. and staphylococcus aureus please it's a request 👍🏼
Do you have a video for preparing xld agar plate? Because xld agar can not autoclave
We will recheck
Do you have a xld agar plate preparing? I don't know why my xld agar always in a liquid

Do you have video on testing of pseudomonas Aureginosal. Please share
We will make it soon
Waiting for part 2.

Good job
Thanks

Thank you so much. I try to find part 2 but cannot find. Please..
With the exception of RVS/MKTT, any proposed salmonella enrichment broth? Thank you
Not at all
Salanite broth
Hi! I just wanna ask if it is necessary to use both RVS and MKTTn for selective enrichment? as well as XLD and BSA for Isolation? thank you so much
Yes. Both needed for selective broth. And you can use only XLD for isolation
Well done my teacher.
Please can I use this method for pharmaceutical samples?
Yes. You can
Is it compulsory to use all the reagents mentioned?
Yes
Mam, what kind of sample you r using?
Animal feed sample
Thanks a lot make more videos for E. Coli, and zeldahl test protein testing and show serial dilution also
Okay. We will make it. Stay with us.
Excuse me, may I ask?
What type of another sample that is easy to detect salmonella ?
Test poultry feces, that has the high possibility to detect salmonella
Pls why did u use 1ml of peptone in mkTTn broth and 0.1ml in RVS broth
Method said so to do that for the selective enrichment of Salmonella spp.
Very useful
Thank you
Mam what sample do you use?
Very info .thanks
It's pleasure.
Thanks for this video ! I couldn't find Part 2, are you going to upload it ? Thanks !

hi your video is very resourceful but can we use selenite broth instead of RVS and MKTTn broth? and also another question, what is the name of this method used? thank you
Yes you can. Method reference is mentioned in the video title
Can we substitute XLD with SS agar
Yes. You can.
Very informative. Can I use distilled instead of peptone water?
No. Peptone water is used for Non-selective enrichment. Non-selective enrichment can not be achieved by distilled water. So you must use Peptone Water.
Thank u so much❤
You're welcome 😊
Thanks! Is part 1 already uploaded?
Both parts are already uploaded.

Can I prepare buffer peptone water from peptone
You can prepare according to the composition of Buffered Peptone Water.
What are the advantages and disadvantages of this method as compared to others detection methods?
1. Advantage: This method involves double enrichment steps which increases detection possibility and facilitates to detect Salmonella more selectively, thus this is the more accurate method compared with other methods.
2. Disadvantage: MKTTN media, frequently, becomes not available in local market and there is no replacement media of it.
Can you name simpler tests for detection of Salmonella with close accuracy?
No other simpler method to be known with close accuracy.
Why does solid foods
need 24hrs for incubation after homogenized with buffered peptone water???..Not only for salmonella detection but also for enumeration of TPC??
24 hours incubation in buffered peptone water is needed for the enrichment of salmonella. But this enrichment is not applicable for TABC or TPC
@@MicroChemsExperiments then what to do for TPC after homogenized with buffered peptone water???
