"Wrap me up like a wagon wheel" refers to the iron band that was hammered around a freshly made wagon wheel to hold the parts together and allow it to cover rough terrain as people travelled across America, as he is doing. And the "Rock me like wagon wheel" is a reference to many obstacles you will drive over and get rocked back and forth before you make it to your destination.

Bob Dylan first wrote What is known today as Wagon Wheel . Boot legged version made the rounds and was named "Rock Me Mama" after a while it became " Wagon Wheel" Old Crow Medicine Show used it and Darius Rucker Recorded it and agreed to royalties to Dylan. It became Number one on Billboards Country charts and a very good showing on Billboards top 100. Love the song It as a certain flair to it that will get you to toe tapping and body motion right from the start.

Maybe we'll have to start calling you 'Wagon-wheel Billy'! 🤣. Anybody want to 'second the motion'?
Have you ever ridden in an old-style wagon, especially in the back? It'll definitely 'rock you'! And if you stand up, it'll rock you right overboard! It's a seriously rough ride, on anything but smooth pavement! Now, on a *good* road, the rocking can be gentle enough to be nice & relaxing, until you hit a real bump.
BTW, did you recognize all the celebrity-folks doing cameos in this?!
I agree with another commenter, watch Bill Engvall's "Where's My Sign?" routine - it'll make you feel better about this!

Wagon is another meaning of the old west of a wagon train where horses pull a wagon. The wagons' wheels rock when they go over rocks, small tree limbs and rough terrain where there are no paths. If the family have a baby, the rocking of the wagon traveling would put the baby to sleep. It is gentle rocking that could put you to sleep. This videos also have Lady Antebellum and Duck Dynasty stars in it.

Presumably a wagon wheel from western stage coach tended to rock or wobble a bit and would rock the coach and make for a bumpy and rocky ride.
I've heard it said that it's referencing a Wurlitzer jukebox where the records where pulled from a wheel shape and dropped onto the player but this song was originally called Rock Me Mama and was only in it chorus form when written by Bob Dylan when he recorded the soundtrack album for the film Pat Garrett & Billy the Kid in the early 1970s. So it's unlikely to have the Wurlitzer connection considering the time setting of the film.
Later in the 2000s Ketch Secor of the Bluegrass/Country/Folk band Old Crow Medicine Show wrote the verses to it. which they then recorded prior to the Darius Rucker cover in this video.
