Gotta be honest, I don't think the mix you are hearing here is what was played live. Watch his solo. He's clearly hitting notes that are not audible. Also watch the official music video and then listen to this one straight after. It is almost identical.

There's also some background vocals that are coming from somewhere else, and at the top you can clearly see them all getting a count in, indicating they're playing to tracks. Doesn't detract from the musicianship on show here, this band is full of world class musicians who are more than capable of pulling off a fantastic live performance, just a production choice for whatever reason.

I'd guess the music is indeed one take. But the shot you are talking about is a video insert from another take, but it's video only. The inserted video is not a perfect match for the music that is of from master audio take, therefore you see the sync error, which shows the differences in performances, visually. This could have been done to cover up some visual mistake, like the camera not in focus, some tech glitching in the video, or some lighting problem. The video insert is covering up something, but they had to suffer the sync error to fix the issue. Most people would never notice the sync problem, but you, sir, have a good eye. I work in pro video, so I can spot it too, but most people wouldn't notice at all. You should be a video editor; that's what editors look for!!!

John Mayer may be the most complete musician package of all time - virtuoso guitar player, excellent singer, but most importantly a song writer on a par with any. On top of that his fan interaction is awesome. We’re going to be hearing his music from now on.
