  • Buffer exchange is an important technique in most macromolecule purification workflows. It can be used to remove unwanted substances such as salts, adjust sample conditions for optimal capture in a chromatography step, or introduce specific components for various assays or sample storage.
    Classical methods for buffer exchange are time-consuming and generate high volumes of waste. In contrast, centrifugal ultrafilters offer easy handling and 90% time savings, while keeping buffer consumption to a minimum.
    When the optimal MWCO is selected, Vivaspin®️, Vivaspin®️ Turbo, and Vivacon®️ will retain your target macromolecule inside a filter unit, while buffer low molecular weight solutes pass through a semipermeable membrane. This technique is commonly used for concentrating macromolecules and can be applied to buffer exchange as well, in a process called diafiltration.
    This tutorial demonstrates the simplicity of discontinuous diafiltration - an iterative process, where the original buffer is removed through multiple rounds of sample dilution and concentration.
