- Is uncooking something possible? In this video I attempt to unboil an egg by using an unlikely ingredient that fully denatures the proteins. Then I dilute it allowing the proteins to refold to their original form. Then I show you how to "cook" an egg without actually heating it by using a strong base to denature the proteins.

As a hairstylist, I thought this was really cool cause this is the exact science used in chemical texturing. Potassium Hydroxide is a common ingredient in a chemical relaxer used to straighten the hair. It's then neutralized to stop the process. Neat to see that eggs react similarly to the disulfide protein bonds in the hair.
It’s not really the “exact science” used in hair relaxes. Here there isn’t a chemical change in the chemical composition of the egg proteins (aka albumin). Like he said the proteins are only being unraveled and the and raveled again. With chemical relaxers the bonds in the protein in hair are being actually being broken and then rebonded which is the actual definition of a chemical reaction.

We do consider it a chemical reaction. There is actually a change to the chemical species as they go from a +dS state to a -dS state. The intramolecular formation of hydrogen bonds, salt bridges, and van der waals interactions contribute a -dH, allowing the reaction to proceed spontaneously under standard conditions. As a result, there are reactants and products other than the peptide sequence itself, but they are thermodynamic values. The "chemical change" doesn't have to do with the elemental constituents of the protein but with the intramolecular bond energies.
Additionally, the denatured protein, which I would classify as unfolded and then coagulated, was kept in the unfolded state by urea which interacts with the hydrogen bonds. Upon dilution these interactions were broken, releasing some free energy (dG) in order to start the process of refolding to the -dS state in an aqueous, buffered environment.

Just a couple of things:
1) It is better practise to put your grams of reagent to add into the flask first and then bring to the final volume with solvent. I'm sure you noticed but 8 M urea requires a lot of urea and will physically occupy a lot of the space in the flask.
2) It is highly unlikely that your renatured egg white proteins (formed upon dilution of urea into water) have folded back into their native state. All you have done is simply dissolved some scrambled proteins in water.
What I love about you is that you are just like mark rober, teaching and inspiring people around the world to get into science but un like mark rober, you teach us more about chemistry while he shares his engineering and physics knowledge.

As a graduate in pharmaceutical chemistry, i can promise you that a denatured protein will NOT go back the way it originally was. Disulfide bonds may re-adhere but they are not guaranteed (in fact it would be highly unlikely) to replicate their original secondary and tertiary structures. Visually this video is cool, but you are not uncooking an egg.

I. Lolly proteins fold with the help of other proteins and go through MANY checks to ensure that they are not misfolded. If a protein cannot fold it gets destroyed in the cell.

Actually, there's not nearly enough urea in piss to do what was shown here, the same way he diluted it with water is how the egg would just stay cooked... And you can't simply let the urine dry out either because of the very high salt makeup as well as any other leftover substances that may have been making their way out of your body... So unless you can filter your piss of all other substances, you might as well just leave your egg cooked
