Kirin Yato's masteries are not significant here as skill level 7 can still one-cycle casters, which is kinda the point of bringing her anyway.
Mostima pot5 is not actually mandatory for this one, however, without pot5 you have to very carefully manage location of phase change. Otherwise none of the pots really matter as long as you have a reasonable amount of pot2's. Myrtle can replace either of the vanguards, just need to allow her to get one more cycle in.

Can Gladiia S2M3 and Ethan S2M3 suffice? I don't have Spalter(My prio to borrow in support unit), and Manticore

To my understanding both are used for week 1, but gladiia starts to suffer in week two due to weight tags. I know CN was still experimenting with gladiia for 7op max risk so it might still be doable but it would be very sketchy. Ethan on the other hand I haven't heard anything about in week 2, but I do know all three ambushers are interchangeable for week 1

You have to shift timings around a bit, but there's plenty of clears that opt for other options. For example, instead of texas alter to take care of the last mortar, there's a clear that uses ebenholz instead
