It is always good if you prepare it fresh, autoclave and use. If you have autoclaved nutrient broth then you can keep it for 1-2week. in refrigerator your cotton plug should Be tight and you should cover it with aluminium foil
If it is dehydrated media powder then you can keep simply it at cool and dry place. Suppose you are a preparing nutrient broth and you have added the water in it then first autoclave it and then you can keep it for one week in refrigerator. Yes, you can keep if it is autoclaved and filled in a sterile bottle under aseptic conditions and you can store it for one week. After one week there is chance of contamination. But I suggest you to prepare it fresh and use fresh.
I have taken 1.5 grams of Nutrient broth powder in a 500 ml capacity flask and in this 500 ml capacity flask I have added 100 ml distilled water. Always make media in Higher capacity flask. For example If you are making 100 ml NB then prepare it in 500 ml capacity flask. It will also leave head space during autoclave and space for oxygen if bacteria are inoculated directly in flask.

why you only added 500mL ??
