    Why 1grm of soil in 50ml water?. Is dis jt an assumption or it's from an SOP?

      Generally soil contains organic materials so it will have large number of bacteria. To get isolated colonies it is better to take 1 gm of soil. It's an assumption. It's not as per SOP

      I have taken 1.5 grams of Nutrient broth powder in a 500 ml capacity flask and in this 500 ml capacity flask I have added 100 ml distilled water. Always make media in Higher capacity flask. For example If you are making 100 ml NB then prepare it in 500 ml capacity flask. It will also leave head space during autoclave and space for oxygen if bacteria are inoculated directly in flask.

    Is this method according to iso? I lately worked in a lab wich they consider an inoculum of 0.1 ml from dilution 1/10 as a1/100 dilution on Petri dish Is that right? I could not understand it because they only spread the inoculum & not diluting it. Please I want an answer.

      We use autoclave to setrilize and dissolve the media. After sterilization we pour the media in petri plate and after agar in plate solidifies we need to keep plates in incubator for drying for 24 hrs -48 hrs. This incubation is also check any contamination while pouring.

      On NA bottle, it is mentioned that for making 1000 ml of NA you need to take 28 grams of NA powder from bottle and suspend it in 1000 ml distilled water. It's just calculation 28 g for 1000ml 2.8g for 100 ml 4.6g for 200 ml Have a great day 😊
