I have one doubt. The nucleotides are added randomly forming different DNA strands. Right? So what if 5'-CG* (bottom 2nd strand) is not formed, only 5'-C* and then directly 5'-CGT* is formed, then accordingly the second nucleotide formed will be considered as "T" instead of "G". Right? But, it will give a wrong reading.

How can you find the strands under the tubes ? From you can identify the strands under the ddNTP strands?
No we don't find strand under the tubes.. we run this sample on agarose gel.. and according to strand length it got separated..
