How if we have the unknown sequence which there's no information about it in database? I mean how can we decide to choose and design the primer for starting the sequencing? An unknown DNA template was sequenced using Sanger’s method. The sequence of the primer used was 5’TCGACGGTGGA 3’. Reactions were carried out in 4 tubes each containing dATP, dGTP, dCTP and dTTP, 32P labelled above primer, DNA polymerase but only one ddNTPs in each tube. For example tube1 has ddGTP, tube2 has ddCTP, tube3 has ddATP and tube4 has ddTTP. Tube 1 produced labeled DNA products of 12nt, 15nt, 18nt and 26nt. Tube 2 produced 14nt, 17nt and 23nt, Tube 3 produced 20nt, 22nt, and 24nt and Tube 4 produced 13nt, 16nt, 19nt, 21nt, and 25nt long products. Write the correct nucleotide sequence of unknown DNA template that should include primer sequence? Can you answer this tomorrow is my assignment deadline please help me
