May I ask why did you not use a cover glass on the slide before viewing it under the scope? Also, could you tell me which stains you used, and would Meth Blue be a good stain to use to check for white blood cells (Leukocytes) in the semen?
I always use cover slips. The only exception is for morphology where I use oil. The stains are described in the WHO lab handbook. Methylene blue is not standard procedure.
Sample issues. Either not all of the ejaculate has been collected or problems in the workflow. Sperm needs a warm place otherwise it will affect sperm motility. It is important to commit to sperm analysis. I always make time for this at the end of the day. There is a certain time slot for doing the analysis. Storing the sample away all day is no option.

why results of different labs varies....how much variation can be seen. If we got tested by two lab in same months...how much percentage variation can be come into see.....
Different people examine samples from different dates with different methods using different equipment. I think, this opens a window into the various possibilities of differing results.
I know this is stupid. But is there any way to collect sperms from water. Like if semen is mixed with water can a viable sperm be extracted out for fertilization (say)?
As per WHO 2010 you should not use vortex and only mix specimen by strile pipette, may I know what kind vortex are you using? Won't it effect the sperms?
I am afraid, that's not true. Thorough mixing is very important to distribute the spermatozoa evenly within the ejaculate in order to avoid incorrect results. A vortex mixer is allowed, in Germany for our regular national proficiency tests it is explicitly recommended. The WHO laboratory manual, 5th edition, 2010 does not ban these devices. It only cautions not to overdo it: "Do not mix with a vortex mixer at high speed as this will damage spermatozoa." (p.18)
Doctor, I had orchitis 30 years ago and developed azospermia. I caught mumps in my late 20's. From then on my sexual appetite has not gone down; so I suppose it has not affected my testoseterone levels, right? Now in my 50's, my libido is still very high, moreso as I lift weights quite regularly. My testosterone is okay despite azospermia right?
I’m really concerned and worried as me and my partner are trying for a baby for the past 10 months no such look yet , but we both have had a megaton of stress ?

Most probably, you didn’t buy a microscope with phase contrast. In usual bright field microscopy you won’t see much.

Yes, absolutely. Without phase contrast , the view is very restricted.

If you mix sperm with water, the survival time of spermatids will be very limited.
