Don't need reservations. There are always "taxis" (pickup trucks with covered benches in the back) rolling through the Settlement and near Sam's restaurant. I can't remember how much it cost but it wasn't much. If you have binoculars, bring them as you'll have the opportunity to see Flamingos as you cross the center of the island but they can be pretty far away.
Last June we rented a safari truck from Anegada Reef Hotel and took the self-guided tour of the island. Plan two nights on the island if you have time on your charter.
