If you will take the time to study DaddyStovepipe's lessons on "Bad Blues","Three Times Lightning" and "Talk with you" first, this one will be a lot easier. Of course a couple of those are not tuned down a half step.

This sound..1:58 . Are you just using the corner of your thumbnail right at the bridge on the d string? It may be your index finger.
I use both thumb and index finger; they pick both the same string - that's how I get that "twangy" sound (at certain places this technique will give you overtones/harmonics).
