One minor correction:
Beta thalassemia unlike alpha is not the result of a a deletion mutation, its a point mutation in the splice site and promoter sequence which results in a reduced synthesis. I've seen this come up as an important distinction between the two types in Q banks.

Just wanted to add a little extra to dirty's amazing video. If they show ringed sideroblast, its only present in the bone marrow whereas on a peripheral smear its called basophilic stippling. How i remember it is stiPPling is in the PeriPhery, SideroBlast are in Bone.
One minor correction:
The PIGA gene does not code for CD 55 and CD 59, It codes for a surface protein on the RBC, which is anchor for these two!
Specifically the GPI anchor protein!
And also, CD 59 inhibits the MAC.

Chronic diseases I know could be under microcytic type of anemia, because of increase in hepcidin level which affects the iron absorption

Recently, I had anemia, or low red blood cell counts, which is common in CLL, and I have CLL.. Red blood cells are needed to carry oxygen to all the cells in the body. With my anemia I experienced fatigue and weakness, and I had pneumonia. My CLL specialist said 'time to treat' as my bone marrow was affected.

At 21:10 you mentioned that C3b causes a MAC to be formed. How does that happen? Is it not C5b and other peptide mediators? That's what it says according to my lectures and Google.
C3b cause it indirectly by initiating the alternating complement pathway and lead eventually to the formation of MAC from c5b and others .

If we have to check b12 or folate deficiency... We can get folate and b12 tests... What's the need of homocystein and mma
B12 can show up as normal even when the patient is B12 deficient. Mentioned in another video :)

So I know the Fenton reaction uses iron to make the hydroxyl radical...
I've always thought the hydroxyl radical was very dangerous because to my knowledge there aren't specific enzymes that scavenge/neutralize the hydroxyl radical and it causes a lot of oxidative damage...
Do you think a deficiency in b6 may exacerbate this hydroxyl radical damage?? Because if you have a lack of b6 then the iron can't be used to make hemoglobin so there would be a lot of iron and a lot of the Fenton reaction and a lot of hydroxyl radical formation and oxidative damage???
Does this make sense is there something about this rationale that isn't not accurate or I maybe missing? I begin med school next year I haven the started yet so I have a lot of holes in my understanding of Biochemistry 😕
If anyone knows or has a deeper insight on these processes I’d really appreciate it.... I know high amounts of iron can apparently accelerate aging? I’m concerned that there’s some hole in my thought processes and there’s something I’m missing or misunderstanding about b6 and iron and Fenton reaction metabolism?
Firstly Fenton Reaction is for free Iron that is circulating in the blood, not the one that is overloaded in mitochondria.After this I think your question becomes self explanatory.
